Objective:Based on the idea of brain-intestinal axis,brain-intestinal interaction,the central position of Atractylodes and peony in IBS model was determined by evaluating the intervention effect of Tongxie yaofang.To reveal the essence of Tongxie yaofang and Atractylodes and peony on protecting spleen and inhibiting liver in IBS model rats,to explain its compatibility principle in modern scientific language,and to explore the relationship between Tongxie yaofang,Atractylodes and peony and brain-intestinal axis and the key points of regulating brain intestinal peptide in IBS model.In order to further clarify the mechanism and key links of the treatment of IBS by inhibiting wood and strengthening earth,the mechanism and key links of the treatment,the law of compatibility of the drug and the compound,the experimental basis for the study of the medicinal properties of traditional Chinese medicine was provided.Methods:The animal model of chronic visceral hypersensitivity of IBS was established by rectal distention stimulation and temporary separation of mother and infant.The visceral sensitivity was evaluated by abdominal withdrawal reflex.To confirm the formation of visceral sensitivity.By observing the general state of IBS rats,measuring the amount of food,measuring the isolated intestinal motility,abdominal withdrawal reflex,open field test,observing the state of Tongxie yaofang and Atractylodes and peony on IBS rats,Effects of absorption and excretion,gastrointestinal motility,visceral sensitivity and animal behavior.The levels of brain-gut peptide CGRP,c-fos protein expression in rat brain,spinal cord and colon tissue were measured by immunohistochemistry.the expression of brain-gut peptide CGRPmRNA,c-fosmRNA in rat hypothalamus,spinal cord and colon by Quantitative Real-time PCR,and the levels of CGRP,c-fos protein expression in rat hypothalamus,spinal cord,colon tissue were observed by Western Blot method.Results:1.After the IBS model was established by rectal distention stimulation,the feeding quantity of the model rats increased slowly(P<0.01),The volume threshold of abdomen lifting and body arching showed decrease(P<0.01),and the movement tension and frequency of the isolated intestinal motility were lower(P<0.01).crossing,rearing,modified times total score of open field test declined significantly(P<0.01),which indicated that the absorption function,gastrointestinal motility function,visceral sensitivity,behavioral index were poor in the model rats.Among them,Tongxie Yaofang and Atractylodes and peony increased significantly(P<0.01),abdominal elevation and back arched volume threshold increased significantly(P<0.01),the threshold value of Tongxie yaofang and Atractylodes and peony increased significantly(P<0.01),and the movement tension and frequency of isolated intestinal motility increased significantly(P<0.01),crossing,rearing movement,modified times was continuously increased on process of administration(P<0.01).2.The expression of CGRP,c-fos in the brain,spinal cord and colon of the model rats was high.The expression of CGRP、c-fos in colon was decreased in different degree(P<0.05),especially in Tongxie yaofang and the group of Atractylodes and peony(P<0.01).Conclusions:1.The effect of Atractylodes-peony and Tongxie yaofang reflected on promoting absorption,regulating gastrointestinal motility,decreasing visceral sensitivity,and regulating animal behavior in IBS model.2.Atractylodes-peony and Tongxie yaofang could decrease the expression level of CGRP,c-fos in the brain,intestine and spinal cord of IBS rats.The mechanism of regulating brain-intestine interaction was revealed from the molecular biological level.3.At the same time,through the biological effect evaluation,the effects of different combinations on the treatment of IBS model rats were as follows:compound group>Atractylodes-peony group>Atractylodes,Group ofpeony,Group of single herbs of Radix Saposhnikoviae,Pericarpium Citri Reticulatae,determine the central position of Atractylodes and peony in Tongxie yaofang.
