OBJECTIVE:To observe the influence of Tongxie YaoFang on the IFN-?and IL-13 of serum in rats with ulcerative colitis(UC),and to identify its un derlying mechanism of action.Methods:70 SD rats of SPF were divided into blank group(10) and module(60), The rats in other groups were treated with enema composed of TNBS with ethanol and suffered from bondage stimulation to establish liver-stagnation and spleen-deficiency syndrome as UC modal.Then were randomly divided into blank group,model group,SASP group and low-,medium-,high-dose of Tongxie YaoFang groups,every model has 10 rats. Except the blank group, UC models were made by means of TNBS/ ethanol enema.The low-,medium-,high-dose of Tongxie YaoFang groups were given Tongxie YaoFang 11, 22,44 g/kg by gavage,SASP group was given 0.3 g/kg,the volume was 10 mL/kg. The blank group and model group were given with the same volume of normal physiological saline for three weeks.Observation rats general situation,do pathological materials for colon after HE staining, IL-13 and IFN-? in rat serum were detected by ELISA.Results:Except the group,model, Then each group have different degrees of inflammatory cells infiltration, hyperplasia of fibrosis and scar repair. The contents of serum and colon IL-13 drugs are rising, Compared with the blank group and model group were statistically significant(P<0.01); Model group IFN- gamma content significantly increased(P<0.01), After treatment, the contents of serum and colon IFN- gamma drug different levels decline(P<0.01)?Conclusion: 1.Experiments have established that bound to stimulate + TNBS and anhydrous ethanol made mold copy to change UC rat model of success.2. UC model rats serum and colon tissue IFN-gamma,IL-13 notable exceptions, Description proinflammatory factor and anti-inflammatory imbalance factor is one of the important mechanisms of the pathogenesis of UC.3. Tongxie Yaofang can significantly drop IFN- gamma and rise the level of IL-13, That the party can adjust the balance between proinflammatory and anti-inflammatory factor, To promote the body's immune balance between Th1/Th2, reduce intestinal inflammation, promote healing of intestinal mucosa ulcer surface, thus achieve the goal of treatment of UC. |
