Secondary Metabolites Of Four Fungi Derived From Soft Corals In The Discovery Of New Drug Lead

Objective: Marine microorganisms have become an important source of bioactive natural products,and have attracted great attention because of novel structures and significant bioactivities.Soft corals,one of the important sources of marine fungi,have become a hotspot among natural products chemists due to that they harbor abundant microbial.In order to find novel lead compounds,this paper investigated the chemical constituents with biological activities of four fungi from soft corals.Methods: The fermented products of four fungi were extracted by acetone and ethyl acetate to afford crude extracts.The crude extracts were then purified by thin-layer chromatography,medium pressure preparative liquid chromatography(MPLC)and semi-preparative HPLC.These isolated structures were identified by nuclear magnetic resonance spectroscopy,mass spectroscopy,and comparison with the reported data.Besides,biological activities of some compounds were be tested.Results: Twenty-one compounds(YN1~YN21)were isolated from Talaromyces sp.SCSIO41201,including diphenyl ethers(YN1~YN11),phenolic acids(YN13,YN14),phenol derivatives(YN16,YN17),one lactone(YN18)and benzene derivatives(YN19~YN21).Seven compounds(YH1~YH7)were isolated from Eupenicillium sp.SCSIO41208,including four griseofulvins(YH1~YH4),one tricholone(YH5)and two benzene derivatives(YH6,YH7).Five compounds(YM1~YM5)were obtained from Eupenicillium sp.DX-SER3(KC871024),including helvolic acid(YM1),nucleotides(YM2,YM3),acetylcholine(YM4)and 4-hydroxybenzaldehyde(YM5).Seven compounds(YO1~YO7)were isolated from Acremonium sp.Z-2-3,including fusidic acid(YO1),ergosterol(YO2),polyphenolic acids(YO3),lecones(YO4)and cyclic dipeptides(YO5~YO7).YN3~YN17 were tested for antifouling activity.YN4,YN8~YN11,YN14 and YN17 showed strong lethal activity against bryozoans,and YN13 displayed weak lethal activities for against bryozoans.YH1~YH4 and YM1 were tested the inhibitory activity of phytopathogenic fungus.Conclusions: All compounds were obtained from these four fungi,including polyketides,peptides,alkaloids,nucleotides,ethers and some benzene derivatives.Among them,three compounds were new and two compounds were isolated from Acremonium for the first time.The anti-fouling effects of YN4,YN8~YN11,YN14 and YN17 were obvious,while,YH1~YH4 and YM1 showed no inhibitory activity.
