| Objective The purpose of this study was to investigate the expression characteristics of BTG3 protein in bladder cancer cells,to further understand its relationship with clinical and pathological characteristics of bladder cancer, and to evaluate the role of BTG3 protein in Patients’ Prognosis. Methods A total of 130 cases of bladder cancer patients who underwent surgical treatment in the department of oncology of our hospital from June 2016 to February 2018 were collected in this retrospective study. The samples were collected from the biological laboratory sample database for centralized time detection. Before the study began, the patients and their families were communicated to clearly inform the relevant process of this study, and the consent was obtained for further study. In this study,paraffin-embedded bladder cancer tissue samples and adjacent healthy tissues of 130 Patients were detected by immunohistochemistry, and BTG3 Protein expression in different tissues was analyzed.Clinical and Pathological indicators of Patients were retrospectively collected through the case management system. After routine follow-up,the follow-up method was combined with telephone follow-up and outpatient follow-up.The Patient’s local recurrence, new lesions(newly found lymph node infiltration or metastasis)were observed. The survival time of the Patients was observed. Results Among the 65 cancer cell tissue samples from bladder cancer patients included in the study,130 patients showed positive expression of BTG3 protein, with a positive rate of 29.23%;BTG3 protein was positively expressed in 40 Patients with para-cancer healthy tissues, with a positive rate of 61.54%.BTG3 protein expression in cancer tissues was significantly lower than that in normal tissues, and the difference was statistically significant(P<0.001).After analysis,we found that BTG3 protein expression was statistically significant when different indicators such as infiltration of bladder cancer tissue,T stage and lymph node metastasis changed.BTG3 protein was positively expressed in37.03%(24/54)of the patients with non-invasive urothelial carcinoma.Among the patients with invasive urothelial carcinoma,33.33%(14/42)had positive expression of BTG3 protein,P=0.019.BTG3 protein was positively expressed in19.05%(4/21)of patients with t1-t2 stage.BTG3 protein was positively expressed in 10.53%(4/38)of t3-t4 Patients(P=0.000).BTG3 protein was positively expressed in 41.94%(13/31)of the patients without lymph node metastasis.Among the patients with lymph node metastasis,14.71%(5/34)had positive expression of BTG3 protein,P=0.016.The difference army has statistical significance. The results of multivariate analysis showed that the degree of cell differentiation and BTG3 expression could be used as independent Prognostic Predictors. The results of cell differentiation were as follows: the risk ratio was 2.243,the 95% confidence interval was 1.421-3.851,P=0.001.The results of BTG3 protein expression status verification were: risk ratio was 2.311,95% confidence interval was 1.131-4.899,P=0.024.Conclusions1.BTG3 protein can be found in healthy cells or cancer cells of the human body, mainly in the cytoplasm of cells, and its expression is relatively high in healthy cells. The negative expression rate of BTG3 protein in healthy paracancer cells of bladder cancer patients was lower than that in bladder cancer tissue cells, and the difference was statistically significant(P<0.05).2.BTG3 protein expression was not significantly correlated with gender, age, histological grade, cutting edge status and other variables.3.BTG3 protein expression was significantly correlated with invasion,T stage, lymph node metastasis and other factors.Univariate analysis showed that low differentiation stage,T stage t3-t4 stage, presence of lymph node metastasis,Presence of infiltration, and negative expression of BTG3 protein all had an impact on the prognosis and survival of patients with bladder cancer. |
