This thesis was consisted of three chapters and summarized the research works during the master’s study.In the first chapter,the chemical constituents isolated from underground parts of C.yunnanensis were introduced.In the second chapter,the analgesic activity of C.yunnanensis was described,the third chapter explores the analgesic mechanism of C.yunnanensis by means of network pharmacology.In addition,and the chemical constituents and biological activities of Cimicifuga from 2016 to now was summarized in the last part.Objective:To study the chemical constituent and analgesic activities of C.yunnanensis.Methods:The compounds were isolated and purified by different skeparation method of column chromatography including silica gel,macroreticular resin,Sephadex LH-20 and preparative HPLC column chromatography.The structure of the compound was identified by physicochemical properties and spectral analysis.The analgesic activity of the isolated compounds was evaluated by the model of acetic acid writhing in mice.Furthermore to explore the mechanism of analgesia through multiple databases related to network pharmacology.Results:Thirty-two compounds were obtained from Et OAc-soluble part and butanol-soluble part of 70% acetone extract of underground parts of C.yunnanensis and identified as 24(S)-3β,24,25-dihydroxy-9,10-seco-9,19-cyclolanost-7(8),9(11)-dien-16,23-dione(1),cimicifuga H-2(2),cimigenol(3),cimigenol-3-O-β-D-xyloside(4),25-Oacetylcimigenol-3-O-β-D-xylopyranside(5),12β-hydroxycimigenol-3-O-β-Dxylopyranoside(6),11β-hydroxycimigenol-3-O-β-D-xylopyranoside(7),12β-hydroxycimigenol(8),25-O-acetylcimigenol(9),actein(10),23-epi-26-deoxyactein(11),27-deoxyactein(12),lupeol(13),β-sitosterin(14),daucosterol(15),cimifugin(16),prim-O-glucosylcimifugin(17),norvisnagin(18),visangin(19),norcimifugin(20), phlorizin(21),ferulic acid(22),isoferulic acid(23),caffeic acid(24),1-feruloyl-β-D-glucopyranoside(25),gallic acid(26),3-hydroxy-3-(4’-methoxyphenyl)propionic acid(27),n-butyl-3,4-dihydroxyphenyllactate(28),bisphenol A(29),3,5-di-O-caffeoylquinic acid(30),monolinolein(31),di-2-ethyl hexyl phthalate(32).The experimental results of writhing analgesia with acetic acid in mice showed that the activity of the compounds 5、10 was better,the analgesic rate of the compound reached 51.17% and 65.82%.Compounds 1,14,16,18,20,22 and the same type of compounds are the main components of the analgesic effect of C.yunnanensis.The analgesic effect of C.yunnanensis achieve analgesic effect by regulating FGF2,ntrk1,F2,MPP2 and other targets.Conclusion:The main components of C.yunnanensis are cimigenol type triterpenoids,acteol type triterpenoids,and flavonoids.It is found that triterpenoids type triterpenoids have good analgesic activity by activity detection,which proves the rationality of the analgesic activity of the plant to a certain extent.
