Expression,Clinical Significance And Mechanism Of Paxillin And P130Cas In Malignant Melanoma | | Posted on:2021-01-27 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Lin | Full Text:PDF | | GTID:2504306128968739 | Subject:Basic Medicine | | Abstract/Summary: | | | 【Objective】To investigate the expression and significance of paxillin and p130 Cas in malignant melanoma(MM),and to explore the inte Ract between two genes.【Methods】1.Eighty malignant melanoma and thirty Intradermal nevus samples were selected and stained with IHC.The expression levels of paxillin and p130 Cas proteins were respectively compared between MM and Intradermal nevus,then the relation of paxillin and p130 Cas with clinicopathological cha Racteristics of malignant melanoma patients was analyzed.2.Thirty MM and eight intradermal nevus in the past 3 years(2017-2019)were selected and detected with Q-PCR.The expression levels of paxillin and p130 Cas mRNA in were also respectively compared between MM and Intradermal nevus.3.Anti-p130 Cas and anti-paxillin siRNAs were constructed and transfected into A375 cells.The interference effect was detected by WB.4.A375 cells were divided into groups A and B.The expression of paxillin was inhibited in group A transfected with anti-paxillin siRNAs,while group B was inhibited the expression of p130 Cas with Anti-p130 Cas.WB and Q-PCR were used to test the interference effect on paxillin and p130 Cas.【Results】1.The positive rates of paxillin and p130 Cas proteins were higher in MM tissues than in intradermal nevus tissues,and the expression of these two proteins was related to gender、metastasis(including lymph node metastasis and extranodal metastasis)and pathological stage in MM patients(P <0.05).There was a significant positive correlation of the paxillin and p130 Cas proteins expression in MM(P <0.05).2.Compared with the intradermal nevus,paxillin mRNA and p130 Cas mRNA were relatively high expressed in the MM group(P <0.05).3.The siRNA interference effect analysis revealed that siRNA had a significant inhibitory effect on paxillin and p130 Cas.4.siRNA interference inhibition test showed that after paxillin gene was knocked out,the protein and mRNA expressions of paxillin were significantly reduced(P<0.05),while the protein and mRNA expressions of p130 Cas were not significantly changed.After p130 Cas gene was knocked out,paxillin and p130 Cas protein and mRNA expression levels were significantly reduced.(P<0.05).【Conclusion】Paxillin and p130 Cas are highly expressed in MM tissues,and the expression of paxillin and p130 Cas is related to the age and metastasis of patients.The expression of paxillin and p130 Cas proteins in MM tissues is significantly positively correlated with each other.There is an interaction between paxillin and p130 Cas.And p130 Cas gene is located upstream of paxillin.Paxillin and p130 Cas can be used as the basis for the differential diagnosis of malignant melanoma,and provide the theoretical basis for their molecular targeted therapy. | | Keywords/Search Tags: | paxillin, p130Cas, malignant melanoma, immunohistochemistry, Real-time PCR, western blot | | Related items |
