Objective: The purpose of this study was to observe the expression of RUNX3 protein in colon polyps of different pathological types and different locations,and to compare the differences with normal mucosa tissues and colon cancer tissue,so as to conduct a preliminary discussion on the role of RUNX3 gene in the occurrence,development and canceration of colon polyps.Methods:A total of 68 adenomatous polyps(including 37 tubular adenomas and 31 villous tubular adenomas),31 serrated polyps,38 cancerous polyps,and 32 normal mucosal tissues more than 15 cm away from the polyps were selected as the control group,all wax blocks of endoscopically resected polyps were collected in the Second Hospital of Dalian Medical University from 2018 to 2020.All the collected cases were diagnosed by pathologists after reading the slides.Using immunohistochemical staining to detect and compare the expression of RUNX3 protein in colon polyps of different pathological types and different locations,and the similarities and differences were observed in combination with the clinical data of the patients.Results:1.RUNX3 protein expressed in each case,and normal mucosa were 90.63%(29/32),adenomatous polyp group were 64.71%(44/68),serrated polyps group were 58.06%(18/31),polyp canceration group were 36.84%(14/38),in the group of normal mucosa and colon polyps and colon polyp canceration of RUNX3 protein expression rate has significant decline.2.Among different pathological types of adenomatous polyps,in tubular adenoma group the positive expression rate of Runx3 protein was 67.57%(25/37),while that in villous tubular adenoma group was 61.29%(19/31),showing no statistical significance.Among the adenomatous polyps with different degrees of dysplasia,the positive expression rates of RUNX3 protein in the low-grade and high-grade dysplasia groups were 73.81%(31/42)and 50.00%(13/26),the positive expression rate of RUNX3 protein was significantly decreased in the normal mucosa group,the low-grade atyphic hyperplasia group,the high-grade atyphic hyperplasia group,and the polyp cancerous group.3.The most common sites of polyps in different pathological types are different.The serrated polyps are more likely to grow in the right colon.The expression of RUNX3 protein in left colon and right colon polyps showing no statistical significance.Respectively compared the right and left colon,the positive expression rate of RUNX3 protein was significantly decreased in the normal mucosa group,the low-grade atyphic hyperplasia group,the high-grade atyphic hyperplasia group,and the polyp cancerous group.4.In gender and age among patients with different pathological differentiation types of colon polyps,the difference showing no statistical significance,and in smoking,drinking alcohol or having hypertension showing the same result.Among the serum related indexes of colon polyps with different pathological differentiation types,only the total cholesterol level of each group was significantly different.The total cholesterol level of normal mucosa group and tubular adenoma group was significantly lower than that of villous tubular adenoma group,serrated polyp group and cancerous polyp group.5.In the positive expression rate of Runx3 protein in different pathological polyps among patients of different genders and ages,the difference showing no statistical significance.Conclusion:1.Runx3 protein expression was progressively decreased in normal mucosa,colon polyps and colon cancer tissues,and Runx3 gene may be related to the occurrence,development and canceration of colon polyps.2.The expression of RUNX3 protein has nothing to do with gender,age,and site of polyp growth,but is related to the pathological type and degree of heterotrophic hyperplasia of colon polyps.RUNX3 gene may become an indicator to predict the degree of pathological changes of colon polyps and the occurrence of canceration in the future.
