Background:In infertility,genitourinary tract infection is one of the important causes.In recent years,more and more evidence has shown that genitourinary tract infections can cause male infertility.Pathogens can cause different degrees of damage to sperm cells.This negative impact can be mediated by the toxins and metabolites secreted by the pathogen,the pathogen directly attaching to sperm cells and subsequent activation of signal transduction pathways related to oxidative stress,apoptosis and inflammation.Lipopolysaccharide(LPS)is the main component of the outer membrane of gram-negative bacteria,and it is also an endotoxin,which can cause a strong immune response and mediate a series of inflammation of the reproductive tract.Levocarnitine(LC)is an indispensable and important component in the β-oxidation process of fatty acids,and as an antioxidant,it can effectively scavenge free radicals,reduce lipid peroxidation,and inhibit apoptosis caused by oxygen free radicals and maintain the stability cell membrane.Objective:To observe the protective effect of LC on the acute injury of the reproductive system of male mice induced by lipopolysaccharide.Methods:Thirty C57BL/6JGpt male mice were randomly and averagely divided into 3 groups.Intraperitoneal injection creates models and administers drugs.Group A:0.9%sodium chloride injection;Group B:LPS solution+LC solution;Group C:LPS solution.After 24 hours,the mice were anesthetized in the abdominal cavity,the eyeballs were removed for blood sampling,and the materials were dissected.The left epididymal tail is used for the assessment of sperm quality(sperm motility,sperm concentration)and the left epididymis and testis are used for histomorphological observation.The serum,right epididymis and testis are used for inflammatory factors(L-1α,IL-1β,IL-6,TNF-α,MCP-1),oxidative-antioxidative stress indicators(GSH-PX,T-AOC,SOD,CAT,MDA),epididymal function indicators(α-GC),inflammation-related pathway molecules(PPAR-y)and fatty acid metabolism pathway molecules(OCTN2,CPT1A,FABP9)detection.Results:1.LC can improve sperm motility and sperm concentration in the acute injury of the reproductive system of male mice induced by LPS;2.Through histomorphological observation,the tissue structure and cell morphology of the epididymis and testis of the mice intervened with LC in advance Although they are injured,the degree of damage is lower than that of mice without intervention;3.LC can reduce the levels of most inflammatory factors in the whole body,epididymis and testis,and increase the expression of PPAR-y;4.100mg/kg LC at 3mg/kg The antioxidant effect of LPS-induced oxidative stress injury in the epididymis of male mice is not obvious;5.3mg/kg LPS-induced acute testis injury in male mice,oxidative stress injury is not dominant;6.LPS-induced males In the acute injury model of the mouse reproductive system,the level of α-GC in the epididymis increases,and LC intervention can reduce the activity and concentration of α-GC,which is correlated with the degree of inflammation.7.LC can increase the expression of OCTN2 and CPT1A,the key molecules of fatty acid metabolism;8.In the acute injury model of male mouse reproductive system induced by LPS,the expression of FABP9 in the epididymis and testis increases,and the intervention of LC can down-regulate its expression,FABP9 There is a correlation with the degree of inflammation.Conclusion:LC can play a protective role in the acute injury of the reproductive system of male mice induced by lipopolysaccharide through regulating fatty acid metabolism and anti-inflammatory.It is the application of LC in the clinical treatment of male reproductive system infections such as epididymitis,orchitis and Epididymis-orchitis,etc. provide a basis.
