| Objective: Through the information mining of various biological information network databases(such as TCMSP database,Gene Cards database,etc.),this study conducted the network pharmacological research of Wenyang Yunpi Decoction for the treatment of peptic ulcer,and obtained the potential active ingredients contained in the formulation of Wenyang Yunpi Decoction and the potential gene targets of this formula for the treatment of peptic ulcer.At the same time,through the construction of PPI network,GO function enrichment analysis,KEGG pathway enrichment analysis and so on,the core targets were screened and the function of potential targets and the participating pathways were predicted,and the “drug-component-target-pathw ay” network was constructed to explore the mechanism of action of this prescription in the treatment of peptic ulcer.Methods: The potential active components of Wenyang Yunpi Decoction and the corresponding drug targets were screened out by data retrieval in TCMSP database using the standard Chinese medicine names or Latin names of each single drug of Wenyang Yunpi Decoction,and then the target names were standardized by Uniprot database for gene names.Then Gene Cards database was searched to obtain peptic ulcer related disease target genes,and the two sets of genes were mapped to obtain the intersection target genes,which were used as the potential targets of this prescription for the treatment of peptic ulcer.Then,the String database and Cytoscape software were used to analyze the potential targets and construct the protein interaction network,and the core targets of Wenyang Yunpi Decoction for the treatment of peptic ulcer were screened out.Then RStudio software was used to conduct the GO function and KEGG pathway enrichment analysis of Wenyang Yunpi Decoction for the treatment of peptic ulcer potential targets in the David V6.8database.Finally,Cytoscape software was used to construct the network of“drug-component-target-pathway”.Results:1.Active ingredients and action targets of Wenyang Yunpi Decoction:Through the TCMSP platform,the screening conditions were set as oral bioavailability > 30% and drug likeness >18.A total of 156 potential active ingredients of traditional Chinese medicine of Wenyang Yunpi Decoction were screened out,and 224 action targets of Wenyang Yunpi Decoction with standardized gene names were identified.2.Target genes and intersection targets of peptic ulcer related diseases:A total of 1001 Peptic Ulcer Ulcer related targets were surveyed in the Gene Cards database and evaluated by “Peptic Ulcer”,“Gastric Ulcer” and“Duodenal Ulcer”.Through RStudio software and R language,the potential targets of Wenyang Yunpi Decoction were mapped to peptic ulcer disease related targets,and a total of 106 intersection targets were obtained.3.PPI network of intersection targets: The String database was used to obtain the data of the protein interaction network of potential targets of Wenyang Yunpi decoction in the treatment of peptic ulcer,and then input into Cytoscape software for the construction of PPI network.There were 105 nodes and 1823 edges in the network.Set the node size and color according to the Degree value,carry out topology analysis on the network according to Cytonca plug-in,and calculate that the double of Degree median is 64,the median of BC is 22.296047,and the median of CC is 0.5745856.The selection condition is that the node’s topology parameters are greater than or equal to the above values.A total of 17 targets were obtained as the core targets of Wenyang Yunpi Decoction in the treatment of peptic ulcer,which were ranked in order of node Degree value as follows: IL6,AKT1,TP53,CASP3,VEGFA,MAPK3,Jun,PTGS2,MAPK8,EGF,MMP9,STAT3,EGFR,CXCL8,CAT,MYC,MAPK1.4.Enrichment analysis results: Through the GO function enrichment analysis and KEGG pathway enrichment analysis of Wenyang Yunpi Decoction for the treatment of peptic ulcer potential targets,a total of 1517 GO entries were obtained,including 1418 biological processes,23 cell components and 76 molecular functions.It involves cellular response to chemical stress,response to molecule of bacterial origin,response to oxidative stress,cellular response to external stimulus,MAP kinase activity,peroxidase activity,and serine hydrolase activity,protein serine/threonine/tyr osine kinase activity,growth factor receptor binding,etc.A total of 131 KEGG pathways were enriched by KEGG pathway enrichment analysis.It involves age-rage signaling pathway in diabetic complications,IL-17 signaling pathway,Endocrine resistance,TNF signaling pathway,TH17 cell differentiation,Apoptosis,FOXO signaling pathway,T cell receptor signaling pathway,PI3K-Akt signaling pathway,HIF-1 signaling pathway,etc.5.Construction of “drug-component-target-pathway” network: The corresponding relationship files of drugs,components,targets and pathways in the data obtained above were input into Cytoscape software for the construction of “drug-component-target-pathway” network,which consisted of 273 nodes and 1587 edges,including 9 kinds of traditional Chinese medicine,128 kinds of active ingredients,106 gene targets and 30 KEGG pathways.It shows that Wenyang Yunpi Decoction in the treatment of peptic ulcer can produce therapeutic effects through multiple components,multiple points and multiple ways.Conclusion(s):1.Through the search and screening of the effective active ingredients of Wenyang Yunpi Decoction,it is found that Wenyang Yunpi Decoction contains a variety of active ingredients.The treatment of peptic ulcer is the result of multiple ingredients,including β-sitosterol,Luteolin,kaempferol,stigmasterol,catechins,etc.are distributed in many Chinese medicines,including anti-inflammatory,antibacterial,and anti-tumor effects,which may be the main treatment of Wenyang Yunpi Decoction for peptic ulcer ingredient.2.Through the prediction of the potential targets of Wenyang Yunpi Decoction in the treatment of peptic ulcer and the screening of core targets,it is found that Wenyang Yunpi Decoction can act on multiple disease targets related to peptic ulcer,of which the main ones are: IL6,AKT1,TP53,CASP3,VEGFA,MAPK3,JUN,PTGS2,MAPK8,EGF,MMP9,STAT3,EGFR,CXCL8,CAT,MYC,MAPK1,and these targets are involved in the process of peptic ulcer cell injury,proliferation,differentiation,repair and the regulation of inflammatory response.3.Through the GO function enrichment analysis of the potential targets of Wenyang Yunpi Decoction in the treatment of peptic ulcer and the results of KEGG pathway enrichment analysis,it is found that the potential targets of Wenyang Yunpi Decoction in the treatment of peptic ulcer are biological functions and signals.The enrichment of pathways is closely related to the regulation of cell proliferation,differentiation,metabolism,apoptosis,inflammation and immune response.4.Based on the analysis of the results of this study,it is found that Wenyang Yunpi Decoction in the treatment of peptic ulcer has an intervention effect on the regulation of inflammatory immune response,the regulation of tumorogenesis,the growth,differentiation and repair of cells in peptic ulcer through the synergistic effect of multi-component,multi-target and multi-pathway,and thus plays a therapeutic effect.At the same time,this prescription may also participate in the immune response to Hp infection,and has a therapeutic effect on Hp infection-related peptic ulcer. |
