Objective:To detect the expression of SOX2、CtBP1 and E-ca in Colorectal adenocarcinoma,Colorectal adenoma and Colorectal tissue,to explore the correlation of the expression of three proteins,and to analyze the relationship between the three proteins and the clinicopathological features and prognosis of CRC,so as to provide new indexes and new ideas for diagnosis,prognosis and individualized treatment of CRC patients.Methods:According to the inclusion criteria,we collected 130 cases of Colorectal adenocarcinoma tissues from January 1,2015 to December 31,2015,which had been undergone radical operation in the Affiliated Hospital of North Sichuan Medical College as the experimental group.Randomly selected 130 cases of Colorectal cancer and 20 cases of Colorectal Colorectal adenoma contrast.Detected the expression of SOX2、CtBP1 and E-cain Colorectal adenocarcinoma,Colorectal adenoma and Colorectal tissue by immunohistochemistry.The comparison of clinicopathological features and protein expression between the experimental group and the control group,and the relationship between the immunohistochemical results of the three proteins and clinicopathological features in the experimental group were tested by chi-square test,continuously corrected chi-square test.Used rank sum test to compare the expression levels of the three proteins in the experimental group and the control group.Used Spearman rank correlation analysis to analyze the correlation among SOX2、CtBP1 and E-ca.The survival data were analyzed by Kaplan-Meier survival analysis and Log-rank test for univariate analysis.Cox proportional hazard regression model was used for multivariate analysis.Used SPSS 21.0 statistical software for statistical analysis of all the experimental data.The inspection level is P<0.05.Results:1.Immunohistochemical staining showed that the positive rate of SOX2 was 84.3%(118/140)in colorectal cancer tissues,70%(21/30)in colorectal adenoma tissues,and 30%(9/30)in normal colorectal tissues.The difference of SOX2 expression in different colorectal tissues was statistically significant(P=0.00).2.The analysis of correlation between SOX2 expression and PTNM stage,patient age,lymph node metastasis,degree of tissue differentiation,and distant metastasis showed statistical significance(P<0.05),while the analysis of correlation between SOX2 expression and patient gender and tumor size showed no statistical significance(P>0.05).3.The positive rate of CtBP1 was 91.4%(128/140)in colorectal cancer tissues,93.3%(28/30)in colorectal adenoma tissues and 93.3%(28/30)in normal colorectal tissues after immunohistochemical staining.There was no statistically significant difference in CtBP1 expression in different colorectal tissues(P=0.902).However,the high expression rate of CtBP1 was 19.3%(27/140)in colorectal cancer tissues,13.33%(4/30)in colorectal adenoma tissues,and 0.00%(0/30)in normal colorectal tissues.Therefore,the expression intensity of CtBP1 in different colorectal tissues was statistically significant(P=0.028).4.The correlation analysis of CtBP1 expression intensity in colorectal cancer tissues with tumor size,lymph node metastasis,patient age,and distant metastasis showed statistical significance(P<0.05),while the correlation analysis of CtBP1 expression intensity with gender,pTNM stage,and degree of tissue differentiation showed no statistical significance(P>0.05).5.Immunohistochemical staining of E-Ca showed a positive rate of 37.1%(52/140)in colorectal cancer tissue,66.7%(20/30)in colorectal adenoma tissue,and 86.7%(26/30)in normal colorectal resection margin.The expression of E-Ca in different colorectal tissues was statistically significant(P=0.00).6.The correlation analysis of E-Ca expression in colorectal cancer tissues with PTNM stage,lymph node metastasis,and distant metastasis showed statistical significance(P<0.05),while the correlation analysis of E-Ca expression with gender,tumor size,patient age,and degree of tissue differentiation showed no statistical significance(P>0.05).7.46 of the 140 patients with colorectal cancer had double positive expression of CtBP1 and E-Ca protein,and 20 had double negative expression.Spearman rank correlation analysis showed that there was no significant correlation between CtBP1 and E-Ca expression in colorectal cancer(r=0.020,P=0.816>0.05),Spearman rank correlation analysis showed no significant correlation between CtBPl and E-Ca expression in colorectal cancer(r=-0.011,P=0.895>0.05).8.There were 110 cases with double positive SOX2 and CtBP1 expression,and 4 cases with double negative expression.Spearman correlation analysis showed that SOX2 and CtBP1 expression were correlated in colorectal cancer(r=0.316,P=0.00<0.05).9.A total of 65 cases of 140 patients with colorectal cancer were visited.By the follow-up date(January 17,2021),20 patients died due to postoperative recurrence or metastasis of colorectal cancer,with an overall survival rate of 69.2%(45/65).The number of patients with SOX2 positive expression was 50,and the postoperative survival rate was 72.00%(36/50).The number of patients with SOX2 negative expression was 15,and the postoperative survival rate was 60.00%(9/15).Kaplan-Meier survival curve showed that the expression of SOX2 in colorectal cancer tissues was not significantly correlated with the survival rate of patients(P=0.317);There were 17 patients with high expression of CtBP1,and the survival rate was 47.10%(8/17);48 patients with low expression of CtBP1,11 of whom died,and the survival rate was 77.10%(37/48).Kaplan-Meier survival curve showed that the expression of CtBP1 in colorectal cancer tissue was correlated with the survival rate of patients(P=0.020<0.05);There were 27 cases with positive expression of E-Ca protein,and the survival rate was 74.10%(20/27),and 38 cases with negative expression of E-Ca protein,and the survival rate was 65.80%(25/38).Kaplan-Meier survival curve showed that E-Ca expression was not significantly correlated with survival rate of patients(P=0.432>0.05);Among the 65 cases of colorectal cancer surveyed,38 had both SOX2 and CtBP1 positive expressions,and the postoperative survival rate was 65.80%(25/38).Kaplan-Meier survival curve showed that the survival rate of patients with both SOX2 and CtBP1 positive expressions in colorectal cancer tissues was not statistically significant different from that of patients with non-SOX2 and CtBP1 positive expressions(P=0.49>0.05).10.Kaplan-Meier survival analysis and Log-rank test were used for univariate analysis of clinicopathological characteristics of 65 patients.Results showed that there was no significant difference in age(P=0.218),sex(P=0.184),tumor size(P=0.672)and prognosis in univariate analysis(P>0.05).Patients with lymph node metastasis and pTNM stage were correlated with survival time.Postoperative survival rate of patients with lymph node metastasis was 46.9%,and that of patients without lymph node metastasis was 90.9%,the difference was statistically significant(P=0.00<0.05);pTNM staging was related to the survival time of patients.Postoperative survival rate was 100.00%in T1-T2 patients and 62.30%in T3-T4 patients,the difference being statistically significant(P=0.018<0.05),distant metastasis was related to the survival time of patients.The postoperative survival rate of patients with distant metastasis was 7.69%,and that of patients without distant metastasis was 84.61%,the difference was statistically significant(P=0.00<0.05).Conclusion:1.The expression of SOX2 is significantly different in colorectal cancer,colorectal adenoma and normal colorectal tissue.The expression of SOX2 is up-regulated in colorectal cancer tissue,and the expression of SOX2 is related to pTNM stage and lymph node metastasis,which may indicate the prognosis of patients.2.The expression of E-Ca is significantly different in colorectal cancer,colorectal adenoma and normal colorectal tissue,and the expression of E-Ca is down-regulated in colorectal cancer,and the expression of E-Ca is correlated with pTNM stage and lymph node metastasis,which may indicate the prognosis of patients.3.CtBP1 was positively expressed in colorectal cancer tissue,adenoma tissue and normal colorectal tissue,but its expression intensity was significantly different,showing high expression in colorectal cancer tissue.The expression intensity of CtBP1 in colorectal cancer tissue was related to the size of the tumor,lymph node metastasis,and the age of the patient.4.The expression of SOX2 and CtBP1 in colorectal cancer tissues is positively correlated,and there is an interaction between SOX2 and CtBP1,which may provide new indicators and new ideas for the diagnosis,prognosis judgment and individualized treatment of patients with middle and advanced colorectal cancer.5.The prognosis of patients with colorectal cancer is closely related to lymph node metastasis,pTNM stage and distant metastasis.
