Radiotherapy is the main treatment for NPC,while the relationship between HIF-1αand GLUT-1 expression in NPC tissues and prognosis of patients with NPC remains to be clarified.Purpose:HIF-1α(hypoxia-inducible factor-1)and GLUT-1(glucose transporter-1)are overexpressed in tissues of patients with nasopharyngeal carcinoma(NPC),and they are closely related to the prognosis of patients with NPC.The purpose of this study was observe the expression of HIF-1αand GLUT-1 in NPC tissues and we can analyse the prognosis of patients with NPC by following up patients for a long time,so as to understand whether the positive expression of HIF-1αand GLUT-1 in patients with NPC are the factor for the long-term poor prognosis of patients with NPC.Method:A total of 86 patients with NPC who were confirmed for the first time by the Pathology Department of Shenzhen People’s Hospital and completed radical radiotherapy were selected.Immunohistochemical assay was used to detect the expression of HIF-1αand GLUT-1 in NPC tissues.In addition,20 cases of patients with chronic nasopharyngeal inflammation confirmed simultaneously were collected as a control.Result:the positive expression rate of HIF-1αand GLUT-1 in NPC tissues was significantly higher than that in nasopharyngeal chronic inflammatory tissue(X2=21.19,P<0.01;X2=17.33,P<0.01).There was no significant correlation between the positive expression of HIF-1αand GLUT-1 in NPC tissues(r=0.039,P=0.719).The positive expression of HIF-1αprotein was significantly correlated with the presence or absence of cervical lymph node metastasis and clinical stage(c2=6.309,P=0.012;c2=5.212,P=0.022).There was no significant difference in 5-year overall survival(OS)between patients with HIF-1αor GLUT-1 positive expression and those with negative expression(P=0.053;P=0.134),while the loco-regional relapse-free survival(LRRFS)of patients with HIF-1αor GLUT-1 positive expression were lower than those with negative expression(P<0.05).The 5-year distant metastasis-free survival(DMFS)of patients with GLUT-1 positive expression was lower than those with negative expression(P=0.035).During the long-term follow-up,the patients with HIF-1αpositive expression had significantly worse long-term survival rate compared with patients with negative expression(P<0.05).The prognostic factor analysis of many clinical factors suggests that the positive expression of HIF-1αand GLUT-1 is an independent factor of poor prognosis in patients with NPC.Conclusion:The expression of HIF-1αand GLUT-1 in NPC tissues can be a prognostic indicator of poor survival in patients with NPC.
