Insomnia seriously reduces people’s quality of life,and insomnia can cause a variety of mental illnesses,including depression,anxiety,etc.Therefore,the research on the treatment of insomnia patients is very important.Currently,drugs targeting GABAa receptors have significant side effects.Therefore,it is necessary to develop a drug with no side effects and a specific therapeutic effect.Current studies have found that multiple G-protein coupled receptors are associated with sleep.At the same time,traditional Chinese medicine is an important means and material basis for our disease prevention,treatment and health care.For thousands of years,traditional Chinese medicine has made great contributions to the prosperity of the Chinese nation.Natural products including Chinese medicine is one of the main source of lead compounds in the development of new drugs.Pharmacological experiments have proved that a variety of traditional Chinese medicines have obvious calming and soothing effects.Including Suanzaoren,Gastrodia elata,Panax ginseng C.A.Mey.,Polygoni multiflori Caulis,Acorus tatarinowii and so on.However,its main mechanism of action and the main sedative and tranquilizing active ingredients are not clear.In this study,the existing dopamine D2/D3 receptor molecular probes in the laboratory were used to trace the components of the dopamine D2/D3 receptors in the.Finally,the dopamine D2/D3 receptor molecular probe was used to detect whether the resulting monomeric compound would act on the dopamine D2/D3 receptor.The main results of this experiment are as follows:(1)Detection of existing dopamine D2/D3 receptor molecular probe stable cell lines in the laboratory.The results prove its effectiveness.(2)Segmenting the Chinese medicine Noctual,and using the segmented part to stimulate the dopamine D2/D3 receptor molecular probe,the results showed that F.3 Fr.4 can be used for dopamine D2/D3 receptors effect.(3)Separation of chemical constituents from Fr.4,9 compounds were isolated and identified,7 steroids: ω-hydroxy emodin(1)Emodin methyl-8-O-β-D-glucoside(2),emodin-8-O-β-D glucoside(3),emodin(6),chrysophanol(7),emodin-6-O-β-D-glucoside(8),Aloe-emodin(9)A chromenone Cassia Cyclohexanone-8-O-β-D-glucopyranoside and a benzene ring derivative,methyl gallate(4).Among them,emodin-6-O-β-D-glucoside was isolated for the first time in the Polygoni multiflori Caulis(4)Using the dopamine D2/D3 receptor molecular probe to detect the activity of the isolated 9 compounds,it was found that several compounds currently screened did not activate the downstream ERK signaling molecule,and the results showed that the ERK phosphorylation level was not in the concentration gradient.Elevated levels indicate that the nocturnal vine can act on the dopamine D2/D3 receptor by other compounds and has not successfully isolated the reactive monomer compounds of the genus.
