Polygoni multiflori Caulis(PMC)is the leafy stem vine of Polygonum multiflorum Thunb.It has been commonly used in Chinese medicine to nourish the heart and calm the mind.There isn’t any research reports on appliance of Drosophila model to detect the improving sleep effects of PMC extracts in domestic and foreign literatures,and the Drosophila model in sleep research has the characteristics of easy breeding,access to a large number of random samples,convenient observation and can promote the accuracy of conclusions from the model aspect,so it is feasible to conduct research on the sleep improvement of PMC with this model.At the same time,the chemical constituents of the extract were isolated and identified,in order to provide some scientific reference for the pharmacodynamic material basis.For the follow up experiment,we obtained the extracts of PMC by means of aqueous extraction and alcoholic precipitation method.In the sleep experiment,when the concentration of PMC reached 0.250%,0.125%and 0.0313%in the culture medium,the effect of PMC extract was to increase the nighttime sleep time of male Drosophila compared to the blank(P<0.01),revealed that the effect of the drug on the nighttime sleep time of male Drosophila worked at lower concentrations;when the culture medium contained 0.500%,0.0625%and 0.0313%PMC extract,the effect of PMC extract on the nighttime sleep time of female Drosophila was highly significant(P<0.01).The extract of PMC had a highly significant effect(P<0.01)on the increase of all-day sleep time of female Drosophila,and when the concentration of the drug was administered at 0.0625%and 0.0313%,it could highly significantly increase the daytime sleep time of female Drosophila(P<0.01),and when the concentration of the drug was administered at 0.500%,the extract of PMC could highly significantly increase the nighttime sleep time of female Drosophila(P<0.01)The effect of PMC extract on the sleep of female Drosophila was mainly in the daytime.From the sleepwake diagram of Drosophila,it was shown that PMC extract could prolong the daytime sleep time of Drosophila and nighttime sleep time of male Drosophila,and significantly reduce the fragmented sleep of Drosophila.The antioxidant capacity of PMC extract in vitro was tested by the DPPH method,and the results showed that the ability of extract to scavenge DPPH radicals(IC50 of 3.06 mg/ml)was slightly weaker than that of VC(IC50 of 0.61 mg/ml).In an antigravity climbing experiment with aged Drosophila,at 0.5%,there was a significant difference between 20-day-old female Drosophila and the blank group,a highly significant difference between 30-day-old male Drosophila,a highly significant difference between 10day-old and 20-day-old male and female Drosophila at 0.25%,and a significant difference between 10-day-old Drosophila at 0.125%,indicating that the fatigue-relieving effect was achieved at low concentrations.The effect of Drosophila melanogaster was shown to be effective at low concentrations.In Drosophila longevity experiment and anti-subacute injury experiment,the survival time of Drosophila melanogaster was longer than that of the blank group.In the experiment of water content in Drosophila melanogaster,the water content in Drosophila melanogaster increased with the increase of administration concentration,which also verified the better antioxidant capacity of the extract of PMC.In this paper,we demonstrated that PMC extract can significantly prolong the sleep time of Drosophila,reduce the fragmented sleep,improve the sleep quality and effectively improve the sleep through Drosophila sleep experiment.It also proved that PMC can relieve fatigue caused by insomnia or fatigue after waking up.Five compounds were isolated from PMC extract by various chromatographic methods,and their chemical structures were identified as malate monomethyl ester-dimethyl ester ether(1),gallic acid(2),caffeic acid(3),methyl gallate(4),and catechic acid(5)with the help of physicochemical identification and spectrums,of which compound 1 is a new natural product.
