| Objective: To assess the value of urinary PCA3 score combined with PCA3-G score,fPSA and %fPSA in diagnosis of prostate cancer.Methods: Both t PSA and fPSA in serum were estimated in 96 patients.Urine were collected for all patients and quantitative real time RT-PCR was used to quantify PSA,GAPDH and PCA3 m RNA.Transperineal prostate biopsy was performed to confirm prostate cancer.ROC(receiver operator characteristic curve)was used to detect diagnostic ability of PCA3 score(103×PCA3/PSA),PCA3-G(103×PCA3/GADPH)score,and combined data sets of various indicators.Results: From 96 patients,51 men were diagnosed with prostate cancer and the rest of patients had BPH or prostatitis.Significant difference between two group patients was observed for serum level of t PSA,fPSA and PCA3-G score.AUC of combined indicators(fPSA,log-transformed PCA3 score and PCA3-G score)was0.79,significantly larger than log-transformed PCA3 score and PCA3-G score.Though combination of indicators improved specificity to 0.864,sensitivity kept unchanged(0.646).Conclusion: PCA3 score(ct PCA3/ct PSAa)combined with fPSA and %fPSA could improve diagnostic rate of prostate cancer in first clinical examination.PSA m RNA level is different between prostate cancer and BPH/prostatitis patients,comparing to housekeeping gene(GADPH).New indicators or assays should be developed for clinical routinely examination for prostate cancer and standard calculating function and proper normalized genes should be advanced for future clinical guidance. |
