Abstract/Summary:
ObjectivesTo investigate the effects of 25 Wei Guijiu pill on uterine index,endometrial thickness,serum antioxidant capacity,blood lipids,serum hormones and estrogen receptor(ER)in perimenopausal syndrome(MPS)rat models the influence of the expression level of subtypes,to explore the regulation effect of Tibetan Medicine 25 Wei Guijiu Pill on rat model of perimenopausal syndrome and its potential molecular mechanism,and provide scientific basis for the biological basis of Tibetan Medicine.MethodsBilateral ovary resection was used to construct MPS rat model.60 female SpragueDawley rats were randomly divided into Sham operation group(Sham),ovariectomized model group(OVX),estradiol group(OVX+E2)and 25 Wei Guijiu pill group(OVX+GJ),with 15 rats in each group.The sham-operation group and ovariectomized model group were given equal volume normal saline,the estradiol group was given estradiol valerate(0.1mg/kg),and the 25 Wei Guijiu pill group was given 25 Wei Guijiu pill(0.36g/kg),for continuous gavage for 8 weeks.The corresponding indexes were detected: 1)Calculate uterine index(UI)and endometrial thickness.2)Detect four items of serum antioxidant capacity and blood lipids.3)ELISA method to detect serum E2,LH,FSH levels.4)Use molecular biology and biochemistry methods to examine the expression of ERα,ERβ m RNA and protein in uterine tissue.ResultsCompared with OVX group,group OVX+GJ index of uterus,endometrial thickness not seen increased(P>0.05),serum level of SOD and CAT activity increased(P<0.05)and MDA activity decreased(P<0.05),TC and TG levels decreased(P<0.05,P<0.01),LDL levels decreased(P<0.01),HDL levels increased(P<0.05),serum E2levels(P<0.01),LH,FSH levels decreased(P<0.05),uterus ERαm RNA and protein expression level there was no significant difference(P>0.05),the expression level of ERβm RNA increased(P<0.01),and the expression level of ERβ protein increased(P<0.05,P<0.01).Conclusions1.25 Wei Guijiu Pill failed to increase the uterine index and endometrial thickness in the MPS rat model,and did not cause endometrial hyperplasia.2.25 Wei Guijiu Pill could increase the activity of T-SOD and CAT in the serum of MPS rat model,and reduce the activity of MDA,indicating that 25 Wei Guijiu Pill could improve the antioxidant capacity of serum in MPS rat model.At the same time,it can reducing the content of TG,TC and LDL and increasing the content of HDL can effectively improve the level of blood lipid metabolism in the MPS rat model,which may be related to the regulation of E2.3.25 Wei Guijiu Pill could effectively increase the level of E2 in the serum of MPS rat models and reduce the levels of LH and FSH,so it is speculated that it may be achieved through the negative feedback of the HPOA axis.4.25 Wei Guijiu Pill could increase the MPS model of rats uterus tissue ERβ m RNA and protein expression,had no obvious effect on ERα,show that the selective estrogen receptor modulators,suggest that the selective estrogen receptor modulators role may be 25 Wei Guijiu pill one of molecular mechanism effectively improve perimenopausal syndrome.
