A Study Of YWHAZ On The Biological Properties Of A549/DDP Cells | | Posted on:2022-05-28 | Degree:Master | Type:Thesis | | Country:China | Candidate:B Y Lu | Full Text:PDF | | GTID:2504306512993869 | Subject:Respiratory medicine | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the effects of YWHAZ on the biological properties of cisplatin-resistant lung adenocarcinoma A549(A549/DDP)cells.Methods: 1.The gene and protein expression of YWHAZ in A549 and A549/DDP cells were detected by q RT-PCR and Western Blotting(WB);the resistance of A549 and A549/DDP cells to cisplatin was detected by CCK8 assay.2.Lentiviruses carrying sh RNA-NC or sh RNA-YWHAZ sequences were transfected into A549/DDP cells to construct cell lines with stable low expression of YWHAZ(sh-YWHAZ)and control cell lines(sh-NC).3.The m RNA expression of A549/DDP cells,sh-YWHAZ cells and sh-NC cells were detected by q RT-PCR;CCK8 assay was used to measure the differences in the sensitivity to cisplatin of the above 3 groups of cells.4.The effects of YWHAZ on the biological behavior of A549/DDP cells were observed by wound-healing assay and Transwell test.Results: 1.The gene and protein expression of YWHAZ in A549/DDP cells were higher than that in A549 cells(P <0.05).The resistance of A549/DDP to cisplatin was higher than that of A549 cells(P <0.05).2.Compared with sh-NC group and A549/DDP group,the expression of YWHAZ in sh-YWHAZ group was decreased at the gene level(P <0.05)and its resistance to cisplatin was also decreased(P <0.05).3.Compared with sh-NC group,cell migration and invasion ability of sh-YWHAZ group were decreased(P <0.05).Conclusion: The expression of YWHAZ was significantly increased in cisplatin-resistant lung adenocarcinoma A549/DDP cells.Knockdown of YWHAZ can reduce the cisplatin resistance of A549/DDP cells,and inhibit its invasion and migration ability,suggesting that YWHAZ plays an important role in the regulation of cisplatin resistance of lung adenocarcinoma cells,which may provide a new direction for the treatment of patients with advanced lung adenocarcinoma in the future. | | Keywords/Search Tags: | A549/DDP, YWHAZ, cisplatin-resistant, migration, invasion | PDF Full Text Request | Related items |
