Objective:To investigate the expression of vascular endothelial growth factor (VEGF) and Neuropilin-2 (NRP-2) in colorectal adenocarcinoma and colorectal adenoma tissues, and their relation with the clinicopathological features of colorectal adenocarcinoma.Methods:The expression of NRP-2 and VEGF were detected by immunohistochemical method (SP method) in 64 cases of colorectal adenocarcinoma, 62 cases of colorectal adenoma, 12 cases of normal mucosa. The sum scores were calculated that were based on the dying depth and the number of posi- tive cells. The score between 0 to3 points was graded into negative, beteeen 4 to 5 points was graded into positive, 6 points or more was graded into strong positive. Expression of NRP-2 and VEGF in colorectal adenocarcinoma, colorectal adenoma, normal mucosa were analyzed using Chi-square test and this association with clinicopathological were explored.Results:1. The positive staining of VEGF protein were mainly located in the cytoplasm. The positive expression rates in colorectal adenocarcinoma, colorectal adenoma and normal mucosa were 81.25% (52/64) 61.29% (38/62) and 25% (3/12). There was significant difference in expression of VEGF between as well as between colorectal adenocarcinoma, colorectal adenoma and normal mucosa (P<0.05). There was significant difference between colorectal adenocarcinoma and colorectal adenoma (P<0.05).2. The expression of VEGF in colorectal adenocarcinoma of clinicopathological features: The expression of VEGF were associated with tumor histological differentiat- ion,infiltration depth, lymph node metastasis and Dukes stages (P<0.05). Expression of VEGF had no correlation to patient's age, sex and tumor site.3. The brown-yellow staining of NRP-2 protein were mainly located in the cytoplasm. The positive expression rates in colorectal adenocarcinoma,colorectal adenoma and normal mucosa were 73.44% (47/64) 37.09% (23/62) and 8.33%(1/12). There was significant difference between colorectal adenocarcinoma and colorectal adenoma (P<0.05). There was significant difference in expression of VEGF between as well as between colorectal adenocarcinoma, colorectal adenoma and normal mucosa.4. The expression of NRP-2 in colorectal adenocarcinoma of clinicopathological features: The expression of NRP-2 were associated with tumor histological differentiation,infiltration depth, lymph node metastasis and Dukes stages (P<0.05). Expression of VEGF had no correlation to patient's age, sex, tumor site.5. Correlation between expression of NRP-2 and VEGF in colorectal adenocarcin- oma: Of 64 patient with colorectal adenocarcinoma, positive expression of both NRP-2 and VEGF were found in 54 cases, negative expression of both NRP-2 and VEGF were found in 4 case , 2 case had NRP-2 positive expression and VEGF negative expression, 4 case had NRP-2 negative expression and VEGF positive expression, Revealed that expression of NRP-2 and VEGF were positively correlated in colorectal adenocarcinoma ( r=0.572, P<0.05).Conclusion:1. Expression of NRP-2 and VEGF were involved in the occurrence and development of colorectal adenocarcinoma. Expression of NRP-2 and VEGF may be associated with degree of malignant in sporadic colorectal cancer.2. The expression of NRP-2 and VEGF was correlated to lymph node metastasis and Dukes stages in colorectal adenocarcinoma.3. Expression of NRP-2 was negatively correlated to expression of VEGF in colorectal adenocarcinoma.4. Expression of NRP-2 and VEGF was correlated to hyperplasia degree in colorectal adenoma.
