| Polygonatum cyrtonema Hua is widely distributed in China.Its quality varies greatly from one to another because of the big difference among different varieties and natural conditions such as climate soils of different origins also vary greatly.In order to study different Polygonatum cyrtonema Hua resources systematically and establish a scientific evaluation system of germplasm resources,we did tests on the heterogeneity of Polygonatum cyrtonema Hua in different places of production,as well as the influence of soil,climatic conditions,etc.on the resources of Polygonatum cyrtonema Hua.And then the typical differential metabolites were screened by researching on the difference of secondary metabolites via metabolomic means.What we resulted would offer reference to distinguish the quality of Polygonatum cyrtonema Hua,as well as to estabish a scientific quality control method for PR medicinal herbs.Here also explains the quality condition in different origin environments.Here comes the main research results as following:1.There are differences in the physiological characteristics of Polygonatum cyrtonema Hua among different origins.The activity of SOD and POD from Anhui is strong,and that of in Fujian is weak.For MDA content,the one from Fujian is high and low from other origins.The plants are tall in plant morphology from three origins as Poyang,Tonggu and Jing’an,Jiangxi Province.The average content of Polygonatum Polysaccharide among the tested 31 wild varieties was 13.53%,which were all higher than the required 7% in Pharmacopoeia of the People’s Republic of China.It is showed in the research that the Polygonatum Polysaccharide content is the highest from Jinzhai,Anhui ones and the lowest from Anhua,Hunan ones.For the content of the total flavonoid content,it showed the highest in ones from Wuyi,Zhejiang and the lowest in Shexian,Anhui Province ones.2.It is found from the relevant analysis that the altitude in ecological factors is the dominant factor affecting the morphological characteristics of PolygonatumPolysaccharide,and the latitude and leaf width are significantly positively related,and the average annual temperature and leaf width are negatively related.There was a significant positive correlation between the Polygonatum Polysaccharide content and the average temperature in July.Also with Polygonatum Polysaccharide content,the number of leaves,the sunshine duration showed a significant negative correlation,accumulated temperature and frost-free period showed a significant positive correlation.Among all the factors related with the Polygonatum cyrtonema Hua content,the average temperature in July was the leading one.The soil of Polygonatum cyrtonema Hua habitat is more to acidic,within which,effective phosphorus and quick-acting potassium are beneficial to the PR plant height,the growth of roots and leaves.3.We studied the formation mechanism of differences within Polygonatum cyrtonema Hua with the help of LC-MS metabolomics technology and multi-statistical analysis.And a total of 25 kind of metabolic difference products were screened,including 5 Amino acid related compounds : Hordatine B,N-Carbamoylputrescine,Agmatine,Triglochinin,Lucuminoside;11 kind of Terpenoids:Alpha-curcumene,Chikusetsusaponin Ia,Cucurbitacin B,Diosgenin,Ent-16-Kauren-19-ol,Gibberellin A5,Monotropein,Nuatigenin,Sclareol,Valtrate,Mascaroside;2 kind of Alkaloids : Vinblastine,3-phenyllactic acid;4Phenylpropanoids compounds:Trans-Cinnamic acid,Coumarin,2-Hydroxycinnamic acid,Sinapic acid;and 2 Flavonoids compounds:Apiin,Glycitin;1 Polyketides:Alpha-Mangostin.The differential metabolites of every compound is something different.Comparing the Polygonatum cyrtonema Hua in different production areas,we can also see the obvious differences,which indicate that the quality of Polygonatum cyrtonema Hua is influenced by the secondary metabolite.Different metabolites can be enriched in 9 pathways,of which 4 pathways have significant effects on the metabolites of Polygonatum cyrtonema Hua from different origins. |
