| Polgonatum cyrtonema Hua,a medicinal and edible perennial plant,is rich in polysaccharides and possesses the funtions of tonifying spleen,nourishing lung,resisting fatigue and protecting cardiovascular.Atherosclerosis is a chronic inflammatory disease with lipid deposition on the vascular wall.However,the effects of gender on atherosclerosis treatment is not fully clarified.Our previous study manifested that polysaccharides exhibited well anti-atherosclerosis efficacy.Accordingly,the current work aims to investigate the intervention effects of Polygonatum cyrtonema polysaccharide(PCP)on atherosclerosis in LDLr-/-mice of different genders and compare the difference of anti-atherosclerosis activity of PCP between male and female mice.PCP(65 and 260 mg/kg/day)was orally administered to high-fat diet(HFD)fed LDL-receptor deficient mice of different genders for 16weeks.The obtained results are as follows:(1)PCP could resist the weight gain and dyslipidemia of LDLr-/-mice caused by HFD.Meanwhile,the effect of PCP on the body weight and dyslipidemia in male mice is more obvious than that in female mice.(2)Effects of PCP on oxidative stress induced by HFD in LDLr-/-mice.Our work found that after PCP treatment,the activity of GSH-PX,SOD in HFD mice were significantly increased,the levels of MDA were decreased.What’s more,the activity of GSH-PX,SOD in male mice were higher than those in female mice,and the levels of MDA were lower than that in female mice after PCP treatment.These findings suggested that PCP could ameliorate the oxidative stress injury,and the amelioration effect of PCP on male mice is better than that of female mice.(3)Effects of PCP on anti-atherosclerotic activity induced by HFD in LDLr-/-mice.The results of oil red O staining of aorta,HE and Masson staining of aortic sinus showed that the aortic plaque area and fibrosis were significantly reduced after PCP treatment,and the male mice had less plaque area and fibrosis than the female mice.These results indicated that PCP could attenuate atherosclerosis,and the intervention effect in male mice was more obvious than that in female mice.(4)PCP could regulate the level of inflammation to relieve atherosclerosis.Quantitative PCR was used to detect the expression of inflammatory factors.The results showed that the m RNA expression of TNF-α,IL-1β,IL-6 were decreased and IL-10were improved in HFD mice after PCP treatment.Moreover,the expression of pro-inflammatory factor m RNA in male mice was lower than that in female mice,and the expression of anti-inflammatory factor was higher than that in female mice after PCP treatment.In addition,the Western Blot results showed that the phosphorylation of P65,ERK1/2,P38,and AKT in HFD mice were decreased after PCP treatment,while the expression of IκB was increased.Furthermore,the phosphorylation of P65,ERK1/2,P38,and AKT in male mice were lower than that in female mice,while the expression of IκB was higher than that in female mice.The above findings reveled that PCP could attenuate the inflammatory response in atherosclerotic mice by regulating NF-κB,MAPK/ERK and PI3K/AKT signals,and the level of inflammation in male mice was lower than that in female mice after PCP treatment. |
