| Objective Because of the different market circulation Polygonati Rhizoma commodity quality,its quality standard is not clear。This situation can not be expected to bring trouble to the safety of clinical medication,so it is urgent to determine its quality evaluation standard,ensure Polygonati Rhizoma safe and effective medication is stable and controllable.Methods ①Through the relevant literature,this paper studied the medicinal history and geographical distribution of Polygonatum cyrtonema Hua.② The 43 batches of Polygonatum were collected to quantify the appearance,such as length,width,impurity,texture and so on.③Referring to the 2015 edition of "Chinese Pharmacopoeia",the items under the Polygonati Rhizoma were tested to see if the alcohol soluble extract,water content,total ash content meet the Pharmacopoeia standards.The contents of elements in As,Cd,Co,Cu,Fe,Hg,Mn,Ni,Pb,Se and Zn were determined by inductively coupled plasma atomic emission spectrometry,to investigate whether it conforms to the standards of "green industry standards for the import and export of medicinal plants and preparations".④ The content of polysaccharide and the content of diosgenin were determined by ultraviolet spectrophotometry and high performance liquid chromatography(HPLC),and whether they met the "Chinese Pharmacopoeia" standard.⑤The common pattern of the glycosides fingerprints of dioscorea zingiberensis was determined by similarity analysis,principal component analysis and system cluster analysis.⑥ The traditional appearance index(impurity)and intrinsic quality index(alcohol soluble extract,water content,total ash content,polysaccharide content and diosgenin content)of the batches were studied by DTOPSIS method,cluster analysis method and principal component analysis method.They were used to comprehensively evaluate the quality of Polygonatum cyrtonema Hua..Results ①Polygonatum cyrtonema Hua.has a wide range of origin.②The length and width of Polygonatum cyrtonema Hua.can not evaluate its quality.③Moisture content of each grade Polygonatum cyrtonema Hua.range from 7.83%to 22.66%,the content of S34 to S35 samples were more than 18%of the Pharmacopoeia,as samples failed,the rest reached Pharmacopoeia standards.Total ash extract content of each grade samples range from 0.27%to 0.86%in line with the Pharmacopoeia.Alcohol-soluble content of each grade samples range from 41.62%to 71.91%,the content of S34 samples was less than 45%of the Pharmacopoeia,as samples failed,the rest reached Pharmacopoeia standards.Determination of the content of the elements in line with the "medicinal plants and preparations for the import and export of green industry standards," the provisions,but S6 lead element far exceeded.④Polysaccharide content of grade samples range from 3.67%to 16.94%,the content of 32,S33,S34,S42,S43 samples was less than 7.0%of the Pharmacopoeia,as samples failed,the rest reached Pharmacopoeia standards.Diosgenin content range from 0.0031%to 0.0242%.⑤A total of 14 characteristic peaks were established,including the peaks 3,4,6,7 and 10,and the contents of these five peaks were crucial for the quality control of Polygonatum cyrtonema Hua..⑥The content of water,total ash content,alcoholic extract,polysaccharide content and diosgenin content play an important role in the quality control of Polygonatum cyrtonema Hua.Conclusion ①There are no obvious real estate areas of Polygonatum cyrtonema Hua.②No processed Polygonatum cyrtonema Hua.should not be used;Length,width,texture can not be used for the evaluation of the quality of Polygonatum cyrtonema Hua.;A small amount of impurities will not affect the quality of Polygonatum cyrtonema Hua..③The contents of Fe,Zn,Cu,Mn and Se can control the quality of Polygonatum cyrtonema Hua.④Longer steaming will reduce the content of polysaccharide polysaccharide,but its diosgenin content increased.⑤The establishment of the fingerprints of the diaprotein fingerprints of Polygonatum cyrtonema Hua,can reflect the types and quantities of chemical constituents.It can comprehensively describe and evaluate the quality of medicinal materials.⑥Through the impurities,moisture,total ash,alcohol soluble extract,polysaccharide content,diosgenin and other traits of Polygonatum cyrtonema Hua,DTOPSIS method,principal component analysis method can be a better evaluation of the quality of Polygonatum cyrtonema Hua. |
