Study On Standardization Of Processing Mongolian Medicinal Aconitum Radix Aconitum Based On Traditional Quality Index

Abstract/Summary:
Objective:In this study,based on the traditional Mongolian medicine and pharmacy theory,modern medicine and pharmacology research methods were applied to analyze the correlation between the slight tongue numb feeling and the connotation alkaloids in the processing of the Mongolian medicine Terminalia chebula and the traditional processing methods,so as to provide reference for the safety of the medicinal Aconitum chebula.Methods:1.Quantitative analysis of alkaloids: HPLC was used to determine the content of Aconitum kusnezoffii in three different Mongolian hospitals.The preparation method of reference substance was to take aconitine standard 1.5mg,hypaconitine standard 0.9mg,neoaconitine standard 5.0mg,benzoylaconitine standard 1.5mg,benzoylhypaconitine 0.9mg,benzoylneoaconitine 5.0mg,and add 10 ml isopropanol chloroform(1:1)mixed solution.The sample preparation method was as follows: 2G of Radix Aconiti kusnezoffii powder was put into a cork conical flask,3ml of ammonia test solution was added,50 ml of isopropanol ethyl acetate(1:1)mixed solution was precisely added,the weight was weighed,and the ultrasonic treatment was carried out(power 300 W,frequency 40KHz;The℃water temperature is below 25 for 30 min,cool,weigh the weight,use isopropanol acetate(1:1)mixed solution to make up the lost weight,shake well and filter.Precisely measure 25 ml of the continued filtrate,reduce the℃pressure below 40 to recover the solvent until dry,precisely add 3ml of isopropanol trichloromethane(1:1)mixed solution to dissolve the residue,tightly plug,shake up,filter,and take the continued filtrate.Take 10 ul of standard solution and sample solution and put them into HPLC for analysis.2.There was slight numbness in the quantitative taste: the Prepared Radix Aconiti kusnezoffii collected by the international Mongolian Medical Hospital of Inner Mongolia Autonomous Region and the raw Radix Aconiti kusnezoffii obtained by the international Mongolian Medical Hospital of Inner Mongolia Autonomous Region were processed by the traditional processing method,each with a diameter of 0.5 cm,and 10 hospital preparation staff were employed to taste them randomly.The size of the processed products,the taste location,the taste time,the generation time of slight numbness and the maintenance time of slight numbness were recorded.3.Acute toxicity test: Acute toxicity test was used to evaluate the acute toxicity of Radix Polygoni.The pre-test measured Dmin and Dmax.There were 60 Kunming mice,half male and half female,with a body weight of20± 2g.They were randomly divided into administration group and control group,with 10 mice in each group.The experiment began with fasting and water withdrawal for 16 h.The control group was given distilled water.1-0.80 dilution,the preparation of 6 kinds of black liquid concentration grass,6 kinds of black liquid concentration grass to concentration of 0.20 ml / 10 g 6 groups of mice by gastric,observe the signs and symptoms.After administration,the symptoms and signs were observed and recorded every 15 minutes for the first 2 hours.The medication was administered for 2-4 hours,and the symptoms and signs were observed and recorded every 30 minutes.The drug was administered for 4-8 hours,and the symptoms and signs were observed and recorded every 1 hour.The drug was administered for 24 hours,once a day was observed,and the symptoms and signs were recorded for 14 days.4.Pharmacological experiments on pain relief and swelling reduction of Radix Aconitum Radix Aconitum in accordance with traditional processing standards:⑴ Eighty SD rats were fed for one week and randomly divided into 10 groups with 8 rats in each group.The model group was given distilled water and the other groups were given the same dose of Aconitum sinensis from different Mongolian hospitals by gavage.Rats in each group were given medicine for 7 days,and the protrusion of the left toe of rats in each group was marked on the 7th day.The left toe of rats was measured by YLS-7B measuring instrument.1 hour after administration,0.1ml carrageenan was injected subcutaneously into the left foot of the rats to establish the model.The left toe of rats was measured at 0.5h,1h,1.5h,2h,4h and 6h after successful modeling.The size difference of the left toe before and after administration was the size of the swelling of the rat toe.The left toe size of the drug administration group and model group was observed and compared at each time period.⑵ Eighty Kunming mice were fed for one week and randomly divided into10 groups.Each group of eight.The model group was given distilled water and the other groups were given the same dose of Aconitum sinensis from different Mongolian hospitals by gavage.The drug was given once a day for 10 days.0.5h after administration on the last day of the experiment,the mice were intraperitoneally injected with 0.6% acetic acid solution 0.10ml/10 g for modeling.After successful modeling,the incubation period and The Times of torsion within 20 min were recorded.The results were as⑴ ⑵follows: Lethal 50%(LD50);Changes in body weight of each group during administration;(3)The swelling of left toe of rats in each group;(4)The number of writhing and incubation period of mice in each group.Result:1.The results of HPLC analysis of Prepared Radix Aconiti kusnezoffii collected from the preparation room of three different Mongolian medical hospitals showed that the reduction rate of total diester alkaloids was as follows: Preparation of Radix Aconiti kusnezoffii in International Mongolian Medical Hospital of Inner Mongolia Autonomous Region >Preparation of Radix Aconiti kusnezoffii in Mongolian Medical Hospital of Alxa League > Mongolian Medicine Radix Aconiti kusnezoffii collected from aruhorqin Mongolian Medical Hospital.That is to say,the loss rate of total diester alkaloids of processed Radix Aconiti kusnezoffii is the lowest in the collection of aruhorqin Mongolian Medical Hospital,and the highest in the collection of international Mongolian Medical Hospital of Inner Mongolia Autonomous Region.The decreasing rate of total diester alkaloids of self-made Radix Aconiti kusnezoffii after collection in preparation room of Mongolian medical hospital was as follows: self-made Radix Aconiti kusnezoffii after collection in aruhorqin Mongolian Medical Hospital > self-made Radix Aconiti kusnezoffii after collection in Alxa League Mongolian Medical Hospital > self-made Radix Aconiti kusnezoffii after collection in international Mongolian Medical Hospital of Inner Mongolia Autonomous Region,Most of them were self-made Aconitum kusnezoffii.It was concluded that the decreasing rate of total diester alkaloids was the highest in the international Mongolian Medical Hospital of Inner Mongolia Autonomous Region,and the lowest in the aruhorqin Mongolian medical hospital.To sum up the above results: the reduction rate of total diester alkaloids in processed Aconitum kusnezoffii made by international Mongolian Medical Hospital of Inner Mongolia Autonomous Region > Aconitum kusnezoffii made by arukorqin Mongolian Medical Hospital > Aconitum kusnezoffii made by Alxa League Mongolian Medical Hospital > Aconitum kusnezoffii made by Alxa League Mongolian Medical Hospital > Aconitum kusnezoffii made by international Mongolian Medical Hospital of Inner Mongolia Autonomous Region > Aconitum kusnezoffii made by arukorqin Mongolian Medical hospital,The loss rate of total diester alkaloids of Radix Aconiti kusnezoffii.That is to say,the loss rate of total diester alkaloids of processed Radix Aconiti kusnezoffii is the lowest in the collection of Mongolian Medicine Radix Aconiti kusnezoffii collected by aruhorqin Mongolian Medical Hospital,and the highest in the self-made Radix Aconiti kusnezoffii collected by international Mongolian Medical Hospital of Inner Mongolia Autonomous Region.Therefore,it has been proved that both the traditional method of processing Radix Aconiti kusnezoffii,the method of baking Radix Aconiti kusnezoffii and the method of processing Radix Aconiti kusnezoffii in Mongolian hospital can remove the toxic components of Radix Aconiti kusnezoffii and achieve the processing effect.2.The traditional standard for processing Terminalia chebula in Terminalia chebula soup is to put 10 kg Terminalia chebula into the soup with 3kg Terminalia chebula boiled in 30 L water,soak in the soup at room temperature for 1 to 3 days,turn it once every two hours,naturally dry,turn it 5-6 times a day.3.Quantitative test with slight tongue tingling sensation: Take 0.5cm of aconite prepared by traditional Terminalia chekhoi decoction and chew it with the incisors for 10-30 seconds.Slight tongue tingling can be felt from the tip of the tongue.The duration of mild tongue tingling is 67.9±41.61 to 99.5±47.18 minutes,and the onset of mild tongue tingling is 1.86±1.19 to 2.48± 1.45 minutes.The slight tongue numbness of Terminalia chebula processed by Terminalia chebula decoction is related to the individual taste.4.The results of acute toxicity test were as follows: the LD50 of Bliss algorithm was 1.198g/kg,and the 95% confidence interval was 0.990 ~ 1.381.According to the acute toxicity grading standard,this sample belongs to grade 3,low toxicity.5.The results of pharmacological experiments on the analgesic and detumescence of Radix Aconitum by the traditional processing method of Terminalia chebula decoction were as follows:⑴ There was no statistical difference in the size of left toe of rats of all groups before the experiment(P > 0.05).Swelling begins 0.5 hours after medication and lasts for 6 hours.After the successful modeling,compared with the model group,there was A tendency of reducing swelling(P < 0.05)in the groups of making Aconitum flutecum B-B,making Aconitum flutecum C-C,making Aconitum flutecum A-A ’,making Aconitum flutecum B-B ’,making Aconitum flutecum C-C ’and baking.After the comparison of toe size,it was concluded that the antiswelling pharmacological effect of Aconitum aconitum A ’group was the best.⑵ compared with the model group,the latency of each group was generally increased(P < 0.05).The shortest incubation period was in the group of Radix Aconiti kusnezoffii and decoction pieces(P < 0.05).The largest number of writhing was self-made Aconitum kusnezoffii.The rate of writhing inhibition was the highest in the self-made Radix Aconiti kusnezoffii group of Alxa League Mongolian Medical Hospital,followed by the self-made Radix Aconiti kusnezoffii group of Inner Mongolia International Mongolian medical hospital.Therefore,we know that the CC ’group has the longest writhing latency and the highest writhing inhibition rate.It is concluded that self-made Radix Aconiti kusnezoffii has the best analgesic effect.Conclusion:Using high performance liquid chromatography(HPLC)method,this study collected in three different Mongolian medicine department of pharmaceutical preparation in hospital kusnezoff monkshood samples and with the traditional processing methods(myrobalan soup cooked up method)homemade six kinds of alkaloid content of a sample,and get the conclusion of the comparison with the traditional processing method of homemade kusnezoff monkshood content than grass low,raw kusnezoff monkshood is higher than that of Mongolian medicine hospital medical system.The results of pharmacological experiments on analgesic and detumescence of Radix Aconitum showed that the pharmacological effects of Radix Aconitum made by the traditional processing method were better.The preliminary conclusion is that in the mouth taste test of processed products with mild tongue tingling sensation,the traditional Terminalia chekhoi prepared by chewing 0.5 cm with the incisors for 10-30 seconds,the slight tongue tingling sensation can be felt from the tip of the tongue,and the maintenance time of the slight tongue tingling is 67.9±41.61 to 99.5±47.18 minutes.The initial time of slight tongue numbness was 1.86±1.19 to 2.48±1.45 minutes,and the slight tongue numbness was related to the individual taste of Terminalia chekhoi.
