Objective: Myrrh is a resin as Burseraceae genus plant from Commiphora myrrh Engl.or Commiphora molmol Engl.and is mainly distributed in South Arabia and North Africa(Somali,Oman,Ethiopia,Egypt,Yemen).As a traditional medicine,it was firstly recorded on the scripture named Treasure of Materia Medica in the Tang and Song dynasties.It has been used to treat arthritis and dysmenorrhea combined with frankincense.Based on the material efficacy of resin not clear,it is worth to explore these chemical and biological components.Our research group has been engaged in the research of traditional Chinese medicine of resin.Previous studies have shown that myrrh has neuroprotective effect and anti-renal fibrosis activities,and have been isolated a series of new sesquiterpene and sesquiterpene dimer compounds.This study mainly adopts a deep exploration on the previous research so as to enrich the structural family of myrrh and provide the scientific basis for clinical medication.Methods: The powdered myrrh(50.0 kg)was extracted with95% Et OH(180 L × 48 h × 3)followed by concentration under reduced pressure to yield a residue(10.0 kg).The residue was dissolved in warm water and partitioned with Et OAc to give the Et OAc soluble part(8.0 kg).After the separation of various chromatographic methods,such as silica gel column chromatography,MCI gel CHP 20 P,RP-18,C-18,Sephadex LH-20,Silica gel,PTLC,and HPLC,series of monomer compounds were eventually separated from the myrrh extract.The structure of the compounds were determined by the analysis of NMR,ECD calculation,and single crystal diffraction.And the new compounds were characterized by physical and chemical data such as ultraviolet,optical rotation,CD,and high-resolution mass spectrometry.At last,some of components were assayed by the biological activities,including antibacterial,anti-inflammatory and lipid-lowering.Results: Totally,31 terpenoids were isolated and identified from Et OAc soluble part of myrrh,of which 9compounds were unpublished.Compounds 1-2 are dinorditerpenoids,compounds 3-5 are sesquiterpenoids dimers,compounds 6-14 are germacrane-type sesquiterpenoids,and compounds 15-18 are guaianese sesquiterpenoids,compounds 19-30 are cadinane-type sesquiterpenoids,and compound 31 is a diterpenoid.Biological activity experiments showed that compound 8 has anti-inflammatory activity,and compounds 28 and 31 have antibacteria activities.Conclusion: New natural products of terpenoids in myrrh were discovered on the research,which enriched the structural types of terpenoids.It was simply screened out for its antibacterial and anti-inflammatory activities,laying a foundation for further in-depth research on the mechanism of myrrh exerting its curative effects and clinical medication.
