Objective Lingzhu Pulvis was used as the research object in this study,and the new structure Lingzhu composite Pulvis was prepared by change the existence form of Powder particle intermediate based on the mechanism of particle design and the characteristics of solvent evaporation method which was realized the optimization of pharmaceutical properties such as quality uniformity and oral compliance of Lingzhu Pulvis.By comparing the changes of pharmaceutical properties of Lingzhu Pulvis,the effect of solvent volatilization-particle design technology on the quality of Lingzhu Pulvis was discussed.And to provide research basis and theoretical basis for the improvement and optimization of the formulation form of traditional Chinese medicine pulvis.Methods 1.Characterization of powder properties of basic materials and extraction of pharmaceutical deficiencies: Fully characterize the powder properties of the basic materials of the prescription,explore the change rule of powder particles in the grinding process,extract the key factors that affect the properties of prescription materials,and excavate the pharmaceutical defects of Lingzhu Pulvis,in order to lay the foundation for the design of particle design technology.2.Determine the introduction mode of particle design,and analyzed the feasibility and technical advantages of modification: Aiming at the pharmaceutical defects of Lingzhu Pulvis,amber-cinnabar-artificial bezoar composite particles were prepared based on the idea of “complementary physical properties” and the design mechanism of powder particles combined with the characteristics of solvent evaporation method.Investigation the powder properties and content uniformity of toxic components of composite particles and physical mixtures to discuss the technical advantages of solvent evaporation-particle design technology in optimizing the quality of traditional Chinese medicine Pulvis preliminary.3.The effect of particle design on pharmaceutical properties of Lingzhu Pulvis: With the compared of the powder performance,oral compliance,quality uniformity and in vitro and in vivo changes of Lingzhu ordinary Pulvis and composite Pulvis,the influence of solvent evaporation-particle design technology on the pharmaceutical properties of Lingzhu Pulvis was explored.Results 1.The powder properties and pharmaceutical deficiencies of basic materials: The physical properties and surface structure of powder particles are closely related to the grinding time.Controlling the particle size of powder particles is the key to control the quality of traditional Chinese medicine powder.By characterization that the Lingzhu Pulvis was showed had poor fluidity and wettability,the artificial bezoar was bitter and fishy in the prescription which seriously affected the oral compliance,and cinnabar was toxic,heavy,and easy to deposit and segregated in the prescription.The above reasons lead to the pharmaceutical defects of Lingzhu Pulvis,such as poor quality,unstable nature,hidden danger of drug use and so on.2.Introduction method of particle design and the feasibility and advantages analysis: The process form of traditional particle design was transformed to achieve in liquid by the mutual movement with particles and realized the structural coating which based on the powder properties of Lingzhu Pulvis.The stable agglomerate structure of the prepared composite particles can be observed under SEM,and the IR also indicated the formation of microstructure.In addition,compared with the physical mixture,the RSD values of chromatism,bulkdensity and toxic contents of the composite particles were significantly decreased,and the taste of artificial bezoar was also significantly improved.The solvent volatilization-particle design technology realized the re-coagulation and combination of multi-component particles with different properties,and significantly improved the pharmaceutics deficiencies of Lingzhu Pulvis,which can be used for the improvement and optimization of the formulation form of traditional Chinese medicine Pulvis.3.The effect of particle design on pharmaceutical properties of Lingzhu Pulvis: The complete prescription regression of amber-artificial bezoar-cinnabar was realized.The results showed that the wettability of Lingzhu Pulvis was significantly improved,the oral compliance was improved,and the RSD of chromatism and bilirubin contents were significantly decreased when comparing the pharmaceutical properties of compound Pulvis and ordinary Pulvis after regression prescription.It suggested that the solvent volatilizationparticle design technology can improve the pharmaceutical properties of Lingzhu Pulvis and provide a reference basis for improving the quality of traditional Chinese medicine powder.Conclusions In this study,a new type of coated particles with stable structure and controllable properties were formed which named Lingzhu composite Pulvis was prepared based on the principle of particle design technology and solvent evaporation methods,and realized the uniform dispersion and Co-coating of multi-component traditional Chinese medicine system with different properties,and the randomly dispersed powder particles were became systematized and regularized.The pharmaceutical defects such as powder properties,oral compliance,mixing uniformity and quality uniformity of Lingzhu Pulvis were improved,and a new breakthrough point was provided for the quality optimization of traditional Chinese medicine Pulvis.
