| Objectives:The preventive effects of SSâ… ,â…¡andâ…¢on atherosclerosis(AS) formation in rats were observed and its possible mechanism was explored, which in order to provide more experimental foundation for the development of anti-atherosclerotic drugs. And the efficacy of SSâ… ,â…¡andâ…¢on atheroselerosis was compared, which could provide an objective preliminary experimental basis for the clinical application of SS ratio.Methods:Rat model with atherosclerosis was established by feeding fatty forage and vitamin D3 (administration vitamin D3 600,000IU/kg by celiac injection at one time) for 12 weeks. 80 healthy male SD rats were randomly divided into 6 groups: 1. normal group ( feed with normal diet); 2. model group (feed with fatty-diet); 3. SSâ… group (feed with fatty-diet+SSâ… :1g/kg.wet.d); 4. SSâ…¡group(feed with fatty-diet+ SSâ…¡:1g/kg.wet.d); 5. SSâ…¢group (feed with fatty-diet+SSâ…¢:1g/kg.wet.d); 6. Simvastatin group(feed with fatty-diet+ Simvastatin:4mg/ kg.wet.d).The SSâ… ,â…¡,â…¢or Simvastatin was fed by gavage to the SSâ… ,â…¡,â…¢and Simvastat in group'mice for 12 weeks, while the normal group and model group were given the same volume of saline as SSâ… ,â…¡andâ…¢groups. After 12 weeks of treatment, the rats were killed, the blood was collected, and the levels of the serum total cholesterol(TC),the low density lipoprotein cholesterol (LDL-C),the high density lipoprotein cholesterol (HDL-C),the triglyceride ( TG), the malonaldehyd (MDA),and the super oxide dismutase(SOD) were measured. The changes in the aortas'ultrastructure were observed through the electron microscope.Results:Comparing with the normal group, serum TC, TG , LDL- C and MDA were significantly increased in the model group (P<0.01); serum SOD in the model group was significantly increased, in the comparison with that of the normal group (P<0. 01) ;the levels of serum TC,LDL-C and MDA in the SSâ… ,â…¡andâ…¢groups were lower than the model group, which had significant differences (P<0.01);The activity of serum SOD in the SSâ… ,â…¡andâ…¢groups was significantly higher than that of the model group (P<0. 01); Serum TG in the SSâ… andâ…¡groups was lower than that of the model group (P<0.01,0.05) ; Serum TG,LDL-C in the SSâ… group were obviously lower than that of SSâ…¡andâ…¢groups (P<0.01,0.05); Serum HDL-C,SOD in the SSâ… group were increased, in the comparison with that of the SSâ…¡group (P<0.05); the contents of TC,MDA in the SSâ… ,â…¡andâ…¢groups had no difference among them (P>0.05). Pathomorphological changes in the aorta: the pathological changes lessened in the SSâ… ,â…¡andâ…¢groups, compared to the model group. The aorta tissue obviously improved.Conclusion:1. Rats model with atherosclerosis were successfully established by feeding fatty forage and vitamin D3 for 12 weeks.2. The SSâ… ,â…¡andâ…¢can improve the metabolism of lipide, such as decreasing the level of TC, TG, LDL-C, and increasing HDL-C notably in the blood.3. The SSâ… ,â…¡andâ…¢can improve the dysbolism of oxygen free radical,strengthen the anti-oxidated capability of the organism, such as increasing the level of SOD,decreasing the level of MDA in plasma.4. Morphological changes may suggest the SSâ… ,â…¡andâ…¢can protect the aorta endothelial cell,improve the pathological change in aorta.5. The SSâ… is more effective that the SSâ…¡andâ…¢. |
