Honeysuckle leaf is the dried leaf of Lonicera Japonica(Caparifoliaceae)plant.Its main chemical components are flavonoids,iridoids,terpenoids and organic acids.It has antiviral,anticancer,antibacterial,anti-inflammatory,and anti-inflammatory properties.Liver,antioxidant,choleretic and other effects It can be used to treat herpes,upper respiratory tract diseases,eliminate free radicals in the body,increase cell activity,anti-aging,hepatitis and liver cirrhosis and other diseases.In this experiment,the anti-inflammatory spectrum effect relationship model of honeysuckle leaf was established for the first time,the effective components were analyzed,the quality standard was studied,and the extract with the best anti-inflammatory effect was selected as the effective components to make anti-inflammatory mask.The main results are as follows:Methods:(1)Prepare 11 different polarity solvent extracts of Lonicera japonicus leaves,examine the chromatographic conditions of Lonicera japonicus leaves different polarity solvent extracts by HPLC,establish their fingerprints,and determine the common peaks of Lonicera japonica leaves with different polarity extracts;analyze the main fragment peaks and molecular ion peaks of each test sample and standard,and compare with the literature to determine the component of each chromatographic peak.(2)Using lipopolysaccharide-induced mouse RAW264.7 cell inflammation model,using the release of 4 inflammatory factors NO,IL-6,TNF-α,IL-1β as detection indicators,study the extraction of different polarities of honeysuckle leaves Anti-inflammatory activity in vitro.According to the gray correlation analysis,the relationship model of anti-inflammatory spectrum effect of honeysuckle leaf was established.(3)Detect various indexes of Lonicera japonica leaves including: moisture,ash content,extract,thin layer chromatography and content determination,etc.,and establish Lonicera japonica leaf quality standards.(4)Using a single factor test method to determine the matrix formula of the mask,and make a mask with honeysuckle leaf extract as the medicinal substance and anti-inflammatory as the main effect.Results:(1)It is determined that the HPLC of extracts of different polar solvents of honeysuckle leaves has 20 peaks in total,and the identified chemical components are chlorogenic acid,luteolin,caffeic acid,rutin,quercetin and luteolin.,Infer 12 known components and 2 unknown components.(2)Anti-inflammatory activity in vitro shows that honeysuckle leaf extracts have good anti-inflammatory activity.According to the gray correlation analysis,the correlation between the peak areas of the 20 common peaks and the anti-inflammatory drug efficacy index is greater than 0.7,indicating that the anti-inflammatory activity of the honeysuckle leaf extract is jointly exerted by multiple components.Among them,the 2 peak deer Spartanin,peak 4 chlorogenic acid,peak 10 truncated loganin,peak 19 quercetin,peak 13 honeysuckle,peak 17 isochlorogenic acid A,peak 9 cryptochlorogenic acid gate The degree is above 0.9,which is the main peak of efficacy.(3)The quality evaluation method of honeysuckle leaf was established: the thin-layer identification study was carried out with luteolin and chlorogenic acid as the control;the microscopic identification showed epidermal cells,stomata,calcium oxalate cluster crystals,etc.;The moisture,total ash,and extract were inspected to determine their reasonable limits;A content determination method with chlorogenic acid,caffeic acid,rutin,luteolin,and quercetin as controls was established.This method can be used to evaluate the quality of honeysuckle leaves.(4)Make honeysuckle leaf extract mask.
