| Objection:To explore the clinical significance of Gasdermin-D(GSDMD)and vascular endothelial growth factor(VEGF)expression in lung adenocarcinoma and their influence on angiogenesis.Methods:To collected clinical pathological data of 80 patients with lung adenocarcinoma who underwent surgical diagnosis and treatment from January 2014 to January 2017 in the Department of Thoracic Surgery of the904 th Hospital of Joint Logistic Support Force of PLA.To detect the expression of GSDMD,IL-1β,VEGF and MVD count(CD34 marker)in lung adenocarcinoma tissue and 30 corresponding adjacent tissues by immunohistochemistry,and calculate the high expression rate,analyze the relationship among GSDMD,IL-1β,VEGF expression and prognosis and the influence on angiogenesis.Results:1.The high expression rate of GSDMD,IL-1β,VEGF and MVD count in the lung adenocarcinoma group were significantly higher than those in the adjacent tissues(P<0.05).2.There is a positive correlation between the expression of GSDMD and IL-1β.And There is a positive correlation among the expression of GSDMD and VEGF and the count of MVD in lung adenocarcinoma(P<0.05).3.Univariate analysis showed that the degree of differentiation,tumor diameter,lymphatic metastasis,TNM staging,GSDMD,VEGF and IL-1β expression levels affected the prognostic survival of patients(P<0.05).4.COX multivariate analysis suggested high expression of GSDMD(HR=1.957,95%CI=1.022-3.748,P<0.05)and high expression of VEGF(HR=2.367,95%CI=1.236-4.532,P < 0.05)are the independent risk factors for patient survival.The degree of differentiation(HR=0.521,95%CI=0.291-0.932,P < 0.05)is an independent protective factor affecting the survival rate of patients.Conclusion:1.The high expression rates of GSDMD,VEGF and IL-1β in lung adenocarcinoma tissues were significantly higher than those in adjacent tissues,and the number of MVD in lung adenocarcinoma tissues was significantly higher than that in adjacent tissues.2.The expression of GSDMD was correlated with tumor diameter,lymphatic metastasis and TNM stage.3.The expression of GSDMD is positively correlated with the expression of VEGF,IL-1β and MVD count.4.The prognosis of lung adenocarcinoma patients are related to tumor differentiation,tumor diameter,lymphatic metastasis,TNM stage,and the expression levels of GSDMD,VEGF and IL-1β in lung adenocarcinoma.5.The high expression of GSDMD and high expression of VEGF are independent risk factors affecting the survival of patients,and the degree of differentiation(moderate and high differentiation)is an independent protective factor affecting the survival of patients.6.The high expression of GSDMD in lung adenocarcinoma may be involved in the formation of tumor blood vessels and tumor progression.In the future,targeting and inhibiting the expression of GSDMD in patients,thereby reducing tumor angiogenesis and delaying tumor growth and progression,may be a new idea for the treatment of lung cancer and help improve the survival rate of lung cancer patients. |
