| The body fluid spots contain relevant information about the ingredients ingested by the donor,which can be used to create a donor profile.Sweat spots are one of the common body fluid spots,and the most common sweat spots at the scene of the case are fingerprints.By analyzing the characteristic components in fingerprints,the donor can be characterized(such as whether he suffers from the hypertension,hyperglycemia and hyperlipidemia),which would provide clues for investigation.This technology has more important significance when individual identification cannot be performed through fingerprints.In this paper,66 common hypotensive,hypoglycemic and hypolipidemic drugs were selected as the research objects,and sweat in fingerprints was used as the matrix.Ultra-high performance liquid chromatography-triple quadrupole composite linear ion trap mass spectrometry technology was used to establish the detection method of 66 drugs in fingerprints.The main research contents were as follows:1.A testing method for 66 kinds of hypotensive,hypoglycemic and hypolipidemic drugs in fingerprints on porous surfaces(filter paper)and non-porous surfaces(glass slide)was established,and an ACQUITY UPLC? BEH C18(3.0 × 100 mm Column,1.7 μm)chromatography column was selected,with 0.01%(V / V)formic acid-water and methanol as mobile phases for gradient elution.Mass spectrometry analysis adopted Scheduled Multiple Reaction Monitoring-Information Dependent Acquisition-Enhanced Product Ion detection method,which could be used to retrieve library researching when high-sensitivity analysis was performed,that could increase the accuracy of qualitative results.The pretreatment method was the protein precipitation.The porous surface was directly cut into pieces and then 0.50 m L methanol was used as the precipitant to extract the drugs;the fingerprint on the non-porous surface was extracted with a cotton swab which was wetted by 50 μL 50%(V / V)methanolwater solution.Then 0.50 m L methanol was used as the precipitant and 10 mg anhydrous magnesium sulfate as a water-absorbing agent.The limits of detection of the 66 drugs were0.001-0.350 ng/patch,the limits of quantification were 0.002-0.500 ng/patch,and the matrix effects were between 78.1%-120%,the recoveries were between 75.5%-119%,the intra-day and inter-day precisions were all within 20%.2.Different deposition surfaces had little effect on the detection of drugs in fingerprints,and this method was applicable to them.The development of fingerprints would reduce the peak areas of the drugs.Among them,the ninhydrin,magnetic powder and “502” glue developing method had relatively little influence.The peak areas of most drugs would decrease with the increasing of deposition time for fingerprints,while some more stable drugs were not affected by this.3.The fingerprints collected from 100 volunteers who took hypotensive,hypoglycemic and hypolipidemic drugs were analyzed by this method,which verified the effectiveness and feasibility of the method. |
