Hair inspection is an important research direction and technical means in the field of forensic identification in our country,and it is also an important content of drug inspection in drug-related cases.Although researches on hair testing techniques and methods are abundant,there are relatively few studies on basic data such as drug content in the hair samples of drug addicts,which is not conducive to the formulation of relevant standards and policies.Therefore,it is urgent to carry out statistical analysis of drug content in the hair of drug users in China.Based on the reality of public security work,this paper selects methamphetamine,heroin and ketamine,three common drugs of abuse as the research objects,and established the ultra-high liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)of 10 common drugs and metabolites in human hair.The cleaning and extraction methods of hair samples were investigated and optimized,and the best pretreatment method was determined.At the same time,this method was used to test a total of 715 hair samples of methamphetamine,heroin and ketamine users.Statistical analysis was carried out on the content data of drugs and metabolites.The main research contents are as follows:Chapter one summarizes the current situation of common drug abuse,the testing methods of common drugs in hair,and related researches on the statistical analysis of drug content in hair at home and abroad,and summarizes and introduces related technical methods and data results.Chapter two established the UPLC-MS/MS detection method of 10 common drugs and metabolites such as methamphetamine.The instrument conditions were optimized and the detection results of 10 kinds of substances were good.In chapter three,the cleaning and extraction conditions of hair samples were investigated and optimized,and the UPLC-MS/MS detection method of 10 drugs and metabolites in hair samples was established.The 10 drugs and their metabolites had good linear relationships(R~2>0.995)within their respective linear ranges.The detection limits and quantification limits were in the range of 0.002-0.02 and 0.005-0.05 ng/mg,respectively,and the recovery range was in the range of 86.73%-106.46%,the matrix effect range was between 80.33%and99.58%,the intra-day precision was less than 10%,and the inter-day precision was less than13%,which can meet the inspection needs.Chapter four examined hair samples from 500 methamphetamine users,101 heroin users and 114 ketamine users.The content ranges of methamphetamine,amphetamine,heroin,O~6-monoacetylmorphine,morphine,codeine,acetylcodeine,ketamine,norketamine and hydroxynorketamine were 0.01-145.35,0.005-15.20,0.01-3.68,0.05-32.72,0.02-14.25,0.02-24.97,0.02-2.56,0.12-73.80,0.03-6.34 and 0.10-58.90 ng/mg.The content ratio of metabolites to original drug showed a certain downward trend with the increase of the original drug content.The cut-off value for methamphetamine,O~6-monoacetylmorphine and ketamine measured by the receiver operating characteristic curve were 0.30,0.68 and 0.55 ng/mg.The above data can provide data reference and support for relevant research and standard formulation to a certain extent.
