| In order to make full use of body fluids to identify the individual characteristics of the suspects,determine the investigative direction during the initial stage of investigation,narrowed down the scope of the investigation.This article aims at physical fluid analysis from chemical analysis.This article selects the common body fluid spot in the crime scene--saliva spot,and establishes an efficient and accurate extraction and detection method for anti-hypertensive drugs in saliva spots,for characterizing saliva donors(whether or not they have high blood pressure).This article selects four representative types of antihypertensive drugs commonly used in the market: Valsartan,Nimodipine,Arotinolol and Indapamide for research.The experimental investigations are as follows:1.The chromatographic and mass spectrometry methods for the detection of Nimodipine,Indapamide,Valsartan,and Arotinolol were established;2.The establishment of the extraction and purification of saliva spots on permeable and non-permeable objects was established;3.Research influences of the object,time,temperature factors and DNA detection on the drugs’ detection;4.The method was applied to the actual sample,verifying the effectiveness and feasibility of the established method.The main results of the study are as follows:1.Use ACQUITY UPLC HSS C18(2.1×50 mm,1.8 μm)as the column;mobile phase organic phase is acetonitrile,aqueous phase is 0.1% aqueous formic acid,use gradient elution procedure and detected by ESI+ source in MRM mode.The parent ion of valsartan was m/z 436.26.Two daughter ions were monitored at m/z 235.14 and 207.15.The parent ion of nimodipine was m/z 419.21.Two daughter ions were monitored at m/z 343.15 and 301.10.The parent ion of arotinolol was m/z 372.11.Two daughter ions were monitored at m/z 298.95 and 316.03.The parent ion of indapamide was m/z 366.03.Two daughter ions were monitored at m/z 132.07 and 91.00.The limit of determination,based on a signal-to-noise ratio of 3:1,Valsartan,Nimodipine,Arotinolo,Indapamide were 0.02ng/mL,0.02ng/mL,0.01ng/mL,0.01ng/mL respectively.This method has a good range of 0.1 ~ 1000 ng / mL,linear relationship,the correlation coefficient is above 0.99.2.The saliva spots on the permeable object(filter paper)were extracted by clipping method,pretreated with methanol precipitation protein method,the recoveries of Valsartan,Nimodipine,Arotinolol,and Indapamide were from 83.1 to 95.1%;Saliva spots on nonpermeable objects(slides)were extracted with cotton swabs and extracted with methanol precipitation protein.The recoveries of Valsartan,Nimodipine,Arotinolol,and Indapamide were between 77.9% and 95.4%.Between the two objects,the intra-day precision and the inter-day precision of the four drugs in the saliva spots were less than 10%,the method had good reproducibility,and the matrix effect was small,so it can be applied to practical cases.3.The results showed that the objects left and the preservation temperature had no effect on the detection.DNA testing had a great influence on the detection of the target.The remaining time only affected the detection of Nimodipine.4.The cigarette butts and cups of three volunteers were collected and tested for practical application,confirming the effectiveness of the method. |
