This article explores the effect of vitamin D uptake combined with aerobic exercise on the expression of PI3K-Akt signaling pathway in the skeletal muscle of type 2 diabetes model mice from the perspective of protein expression.During the experiment,the routine indicators of mice were strictly tested.After the experiment,the mouse blood and tissue samples were extracted and the corresponding indicators were tested.And study the mechanism of vitamin D combined with aerobic exercise to improve insulin resistance,so as to provide the corresponding theoretical basis and application reference for the prevention and treatment of type 2 diabetes.Objective:(1)To study the effect of vitamin D combined with aerobic exercise on insulin resistance in db / db diabetic mice.(2)To explore the mechanism of vitamin D combined with aerobic exercise on PI3 K / Akt signaling pathway in db / db diabetic mice.Provide the corresponding theoretical basis and application reference for the prevention and treatment of type 2 diabetes.Method: 32 male 7-week-old male db / db mice(SPF grade)were fed after one week of adaptive feeding.Blood samples with a fasting blood glucose of not less than 16.7 mmol / L were eligible for type 2 diabetes(T2DM).After numbering the mice,they were divided into 4 groups according to the random number table method,with 10 mice in each group.The control group was given intraperitoneal injection with equal amount of soybean oil three times a week.The vitamin D group was also given high-fat diet and intraperitoneal injection at a dose of 7ug / kg of vitamins.The exercise group performed moderate-intensity treadmill aerobic exercise five times a week,and the vitamin D combined exercise group performed treadmill exercise every Friday At the same time,he was given an intraperitoneal injection of vitamin D three times a week for 8 weeks.After the experiment,the mice were fasted for 12 hours,water was not allowed,blood was collected from eyeballs,serum was separated,and anesthetized,and the skeletal muscles and related tissues were put into quick freezing in liquid nitrogen and stored in-80 ° C refrigerator.German EKF blood glucose lactate analyzer was used to measure blood glucose,and microplate reader was used to detect blood lipid levels.Insulin and vitamin D were measured using Elisa kit.Immunohistochemistry and Western Blot were used to detect liver and skeletal muscle tissues,and the expression level of related protein kinase B(Akt)in PI3 K /Akt signaling pathway was detected.Result:(1)Compared with the control group,the weight,blood glucose,and insulin resistance index(HOMA-IR)of the experimental group all decreased(P <0.05).(2)The levels of blood lipid index TG,TC,HDL-C and LDL-C all decreased.(3)After intervention of vitamin D and aerobic exercise,the liver cell morphology of mice in each group showed a certain degree of improvement.(4)WB results showed that compared with the control group,the expression of Akt protein in skeletal muscle of vitamin D group,exercise group,vitamin D +exercise group all increased to varying degrees,and the expression of Akt protein in skeletal muscle of vitamin D + exercise group increased significantly.Compared with the control group,the Akt protein content in the liver tissue was higher in the intervention group,and the difference was statistically significant(P<0.05).Compared with the control group,the exercise group and VD + exercise The Akt protein in the group increased,and the difference was statistically significant(P <0.01).(5)Immunohistochemical results showed that compared with the control group,the expression of Akt protein in the skeletal muscle tissue of the intervention group increased,of which the exercise + VD group had the highest expression,followed by the exercise group,and the VD group had the least.The results showed that the protein expression of Akt in the VD + exercise group was the most obvious.Compared with the control group,the expression of Akt protein in the skeletal muscle tissue of the intervention group was increased.The expression level of the exercise + VD group was the highest,followed by the exercise group,and the VD group was the least.The results showed that exercise and VD intervention in the VD + exercise group enhanced Akt protein expression in skeletal muscle of mice.Conclusion:(1)Vitamin D combined with aerobic exercise reduces LDL-C and FINS,improves insulin tolerance and OGTT in type 2 diabetic db / db mice,reduces the insulin resistance index,and significantly improves insulin resistance.(2)Vitamin D combined with aerobic exercise reduces the TG content of type2 diabetic db / db mice and improves liver fat-like changes.(3)The mechanism of vitamin D combined with aerobic exercise to improve the IR of type 2 diabetic db / db mice may be through the PI3 K / Akt signaling pathway in the liver and skeletal muscle to promote the absorption and utilization of glucose by the liver and muscle tissue of mice Thereby increasing the use of glucose by muscles,promoting the synthesis of glycogen in the body,regulating the homeostasis of blood sugar in the body,improving glucose metabolism and IR.
