Objective::Taking the KM mice as the research object,this topic is to study the expression and effects of related cytokines of aerobic exercise on the PI3K/AKT/GSK3? signaling pathway of chronic cerebral ischemia mice whose right carotid artery is ligatured.Methods:Take the chronic cerebral ischemia mice model built by Yoshizaki as a reference,select KM mice as the experimental animal,and then establish a chronic cerebral ischemia mice model.After establishing the model,randomly divide the mice into several groups,such as the Blank Group(CG),the Sham Operation Group(SO),the Model Group(M),the Swimming Group(SW),and the Treadmill Group(TM),10 in each group.Each Intervention Group intervenes aerobic exercise on the third day after establishing the model.Neuroethology Assessment will observe and record the sensorimotor behavior of the mice after establishing the model and after intervention.After the experiment,let the mice fast overnight and separate the brain tissue and draw some materials.Morphology will observe the distribution of the nissl bodies of the mice.The Immunity Group will inspect the expression of protein of PI3 K,Akt,GSK3?,MCL-1 in the signaling pathway.Observe the effects of aerobic exercise on the PI3K/AKT/GSK3? signaling pathway of chronic cerebral ischemia mice,and then discuss its neuroprotective effect on chronic ischemic brain injury mice.Results:(1)Neuroethology Assessment: After establishing the model,the neurobehavior of the mice showed no significant difference.After the exercise intervention,the score of the Exercise group was lower than that of the Model Group(P<0.05),and the scores of the Blank Group and the Sham Operation Group were the lowest,and there was no significant difference between these two scores(P>0.05).(2)Result of the nissl's staining: there was a large number of big nissl in the Sham Operation Group,arranged closely.The nissl showed pleomorphism,and the neurons suffered little damage,reflecting the strong function of nerve cells in synthesizing protein and transmitting information.The Model Group arranged sparsely and irregularly,and the quantity was small,the neurons suffered more damage.Compared with the Model Group,the quantity of nissl in Swimming Group was bigger,and arranged closely(P<0.05).But there was a great difference when compared with the Blank Group(P<0.05).The quantity of nissl in the Treadmill Group was bigger than that of the Model Control Group(P<0.05),ranked second only to the Sham Operation Group,with a large number of neurons,arranging regularly.(3)The result of the immunohistochemistry showed: the positive expression of PI3 K,AKT,MCL-1 increased obviously in the Aerobic Exercise Group compared with the Model Group(P<0.01),which was the highest in the Treadmill Group.Compared with the Blank Group,the there was less expression of protein(P<0.05).The expression of GSK3? in the Swimming Group and the Treadmill Group was the lowest,which showed significant difference compared with the Model Group.Compared with the Swimming Group,there was less expression in the Treadmill Group,with statistical significance(P<0.05).Conclusion: Aerobic exercise plays a role in brain protection for chronic ischemic brain injury mice,enhancing the endurance capacity of the brain tissue to cerebral ischemia.The effects of the Treadmill Group is better because Aerobic exercise promotes the related factors of blood vessel,nerve regeneration and restoration in the PI3K/AKT/GSK3? signaling pathway,reduces the death of neurons caused by cerebral ischemia,and plays a role in brain protection.
