| Objective: With the improvement of people’s living standards and the advent of an ageing population,more and more women are entering the Perimenopausal period.During this special stage,women’s various functions change from exuberant to aging,and they will experience physical,psychological and other changes.It is very easy for women to have different degrees of emotional disorders.If they are not detected and adjusted in time,it will eventually lead to the occurrence of Perimenopausal depression.Perimenopausal depression refers to a type of mental illness with persistent low mood,memory loss,slow thinking and cognitive function decline that occurs during the period from ovarian function decline to 1 year after Perimenopausal sexually transmitted diseases.Depression is a series of symptoms that make people miserable,seriously affect women’s daily activities and physical and mental health,and become an important factor in reducing the quality of life.In recent years,it has been clearly recognized that physical activities can promote physical and mental health,participate in emotional regulation are easily accepted by patients,and have significant long-term effects.Therefore,this study observed the effects of moderate-intensity treadmill exercise on Perimenopausal depression-like behavior,Hippocampal inflammatory factors interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis The effect of factor-α(TNF-α)levels and the expression of apoptosis factors Bcl-2,Bax,caspase-3 protein levels,to explore the possible mechanism of exercise in the treatment of Perimenopausal depression,for the future exercise exercise on the Perimenopausal period The improvement of depression provides a theoretical basis.Methods: 40 female SD rats were randomly allocated to 4 groups,which included sham-operated rats(SHAM),ovariotomy rats(OVX),stressed OVX rats(OVX+S)and stressed OVX rats treated with exercise(OVX+S+EX).After the exercise,the depression-like behaviors of the rats in each group were tested by the sugar water preference test,the forced swimming test and the open field test;the hippocampal inflammatory factors interleukin-1β(IL-1β),interleukin-6(IL-6)The content of tumor necrosis factor-α(TNF-α),and the protein levels of Hippocampal apoptosis factors Bcl-2,caspase-3 and Bax were detected by western blot.Results:(1)Compared with the SHAM group,in the sugar water preference test,the sugar water consumption and sugar water preference rate of the OVX group were significantly decreased(P<0.05),but the total fluid consumption had no significant difference(P>0.05);In the sugar water preference test,the sugar water consumption and the sugar water preference rate of the rats in the OVX+S group were significantly decreased(P<0.01),and the total fluid consumption was not significantly different(P>0.05).In the forced swimming test,the immobility time of the rats in the OVX group was significantly increased(P<0.05),but there was no significant difference in the climbing time(P>0.05),the immobility time of the rats in the OVX+S group was significantly increased(P<0.01),and the climbing time was significantly decreased(P<0.01);In the field test,the residence time and fecal particles in the central grid of the rats in the OVX group increased significantly(P<0.05),and there was no significant difference in the number of standing,crossing grids and modification times(P>0.05),in the OVX group,the dwell time and fecal pellets increased significantly(P<0.01),while the number of crossing grids,standing times and modification times decreased significantly(P<0.01).Compared with the OVX group,in the sugar water preference test,the sugar water consumption and sugar water preference rate of the OVX+S group were significantly decreased(P<0.01),but there was no significant difference in the total fluid consumption(P>0.05).The immobility time of the rats in the OVX+S group was significantly increased(P<0.05),and the climbing time was significantly decreased(P<0.05 or P<0.01),the number of crossing grids,the number of standing,and the number of modification were significantly decreased(P<0.05 or P<0.01).Compared with the OVX+S group,in the sugar water preference experiment,the sugar water consumption and the sugar water preference rate of the OVX+S+EX group were significantly increased(P<0.05 or P<0.01),but there was no significant difference in the total fluid consumption(P>0.05);in the forced swimming test,the immobility time of the OVX+S+EX group was significantly reduced(P<0.05),and the climbing time was significantly increased(P<0.01);in the open field test,the OVX+S+EX group The residence time in the central grid and the fecal particles were significantly decreased(P<0.05 or P<0.01),and the number of modification,crossing grids,and standing times were significantly increased(P<0.05 or P<0.01).(2)The inflammatory factors in the hippocampus of the rats in each group were detected by ELISA.Compared with the SHAM group,the levels of inflammatory factors IL-1β and IL-6 in the hippocampus of the rats in the OVX group were significantly increased(P<0.05),but there was no significant difference in TNF-α(P>0.05),in OVX+S group IL-1β,IL-6 and TNF-α were significantly increased(P<0.01).Compared with the OVX group,the IL-1β,IL-6 and TNF-α of the OVX+S group were significantly increased(P<0.01).Compared with the OVX+S group,the IL-1β,IL-6 and TNF-α of the OVX+S+EX group were significantly decreased(P<0.05).(3)The apoptosis factors of Hippocampal cells in each group were detected by western blot.Compared with SHAM group,the relative expression of Bcl-2 protein in OVX group was significantly decreased(P<0.05),while the relative expression of Bax and Caspase-3 protein was not significant(P>0.05).The relative expression of Bax and Caspase-3 protein in OVX+S group was significantly increased(P<0.01),and the relative expression of Bcl-2 protein was significantly decreased(P<0.01).Compared with the OVX group,the relative expressions of Bax and Caspase-3proteins in the OVX+S group were significantly increased(P<0.01),and the relative expression of Bcl-2 proteins was significantly decreased(P<0.01).Compared with the OVX+S group,the relative expressions of Bax and Caspase-3 proteins in the OVX+S+EX group were significantly decreased(P<0.05),and the relative expression of Bcl-2 proteins was increased(P<0.05).Conclusions:(1)In this experiment,ovariectomy combined with chronic unpredictable stress was used to establish a rat model of Perimenopausal depression.It was confirmed by behavior that the rat model of Perimenopausal depression was basically successful.(2)The Perimenopausal depression rat model established by ovariectomy combined with unpredictable stress can lead to a decrease in the ability of the rat to explore,a decrease in the degree of curiosity about the fresh environment,depression-like behaviors such as anhedonia and increased despair,and hippocampus-like behaviors.The levels of inflammatory factors and apoptotic factors were abnormal.Therefore,it is speculated that the depressive behavior of Perimenopausal depression rats may be related to the inflammatory response and apoptosis of brain tissue caused by ovariectomy and chronic stress.(3)4 weeks of moderate-intensity treadmill exercise can improve depression-like behaviors in Perimenopausal depressed rats,and reduce Hippocampal inflammatory response and apoptosis,suggesting that hippocampus may be one of the brain regions that exercise interferes with Perimenopausal depression.And the improvement of Perimenopausal depression in rats by exercise may be related to the reduction of inflammatory response and inhibition of apoptosis,thereby playing a role in brain protection.(4)Exercise can be used as an alternative or adjunctive treatment for Perimenopausal depression,but the long-term effects of exercise need further research. |
