Objective:Through the different exercise duration and exercise load swimming exercise,we can understand the changes of testosterone and inflammatory factors in rats.To explore the effect of prolonged and heavy exercise may affect the secretion of testosterone in rats by affecting inflammatory factors.In order to reasonably arrange exercise time and exercise load,scientifically formulate training plans to speed up fatigue elimination and improve athletes'function level,and provide theoretical basis.Methods:48 healthy male Sprague-Dawley rats were randomly divided into 4 groups:normal control group(C,n=12),60 min exercise group(CEA,n=12),120 min exercise group(CEB,n=12),joint weight-bearing exercise for 60 minutes Group(SE group,n=12).Feeding methods:Ordinary feed feeding.Exercise program:There was no exercise intervention in group C,CEA group and CEB group used weightless swimming exercise intervention,swimming duration was 60min and 120min per day;SE group used joint weighted swimming exercise,weight was 6%of body weight,swimming duration was 60min per day.Each group of rats was exercised for 6 days per week and rested for 1 day for a total of 10 weeks.Before the experiment,the rats in the exercise group performed adaptive swimming for 1 week.After exercise intervention,serum testosterone(T),leptin(LEP),nuclear factor-?B(NF-?B)and tumor necrosis factor-a(TNF-a)were detected by enzyme-linked immunosorbent assay(ELISA).Interleukin-6(IL-6),monocyte chemoattractant protein-1(MCP-1),C-reactive protein(CRP),and testosterone(T)levels in testis;The qRT-PCR method was used to detect the mRNA expression of TNFR and IL-6R in the testis.HE staining was used to observe the morphological changes of the testis.Results:(1)There was no significant difference in body weight between the groups before the start of swimming training;the body weight of the rats showed a downward trend after exercise training,and compared with the control group(C),60 minutes exercise group(CEA)and 120 minutes exercise group(CEB)There was a significant difference between the 60-minute exercise group(SE)group and the combined weight-bearing group(P<0.01).(2)After 10W swimming training,the spermatogenic cells of the testis of group C and CEA were arranged in order.The arrangement of spermatogenic cells in the seminiferous tubules of the testis of the CEB group and the SE group was disordered.(3)Different exercise duration and exercise load can lead to a decrease in serum T levels in rats,compared with C group,CEB group decreased especially significantly(P<0.01).Compared with CEA group,serum T level in SE group was significantly lower(P<0.01).(4)Different exercise duration and exercise load can increase the levels of serum LEP and NF-?B in rats.Compared with group C,the level of serum LEP in CEA rats increased significantly(P<0.05),but there was no significant difference in NF-?B levels.Changes in serum levels of LEP and NF-?B were significantly higher in CEB group(P<0.01).Serum levels of LEP and NF-?B were significantly higher in SE group than in CEA group(P<0.01).(5)Compared with group C,serum TNF-a and IL-6 levels in CEA rats decreased significantly after 10W swimming training(P<0.05),while CEB group increased significantly(P<0.01).Serum levels of TNF-a and IL-6 were significantly higher in the SE group than in the CEA group(P<0.01).(6)Compared with C,CEA rat serum CRP,MCP-1 levels did not change significantly,but CEB group serum CRP was significantly increased(P<0.01),MCP-1 was significantly higher(P<0.05);SE group and CEA Compared with the group,serum CRP and MCP-1 levels increased significantly(P<0.01).(7)Different duration and exercise load can lead to a decrease in the T level in the testis of rats,in which the CEA group significantly decreased(P<0.05),and the CEB group decreased especially significantly(P<0.01).Compared with CEA group,the T level in testis decreased significantly in SE group(P<0.01).(8)The results of qRT-PCR showed that the duration of exercise and exercise stress could increase the mRNA levels of TNFR and IL-6R in testis of rats.Compared with group C,the mRNA levels of TNFR and IL-6R in testis of CEA group had no significant difference.In terms of sexual changes,the mRNA levels of TNFR and IL-6R in the testis of the CEB group were significantly higher than those of the CEA group(P<0.01).Compared with CEA group,the levels of TNFR mRNA and IL-6R mRNA in testis of rats increased significantly(P<0.01).Conclusion:(1)Reasonable exercise duration can slow down the body's inflammatory state.Excessive exercise duration and exercise load can cause inflammation.(2)Excessive exercise duration and exercise load can cause changes in serum leptin,serum inflammatory factors,and expression levels of related inflammatory factor receptors in the testis,as well as affecting testicular tissue morphology and affecting testosterone in serum and testis.s level.
