Polygonatum cyrtonema Hua.Is a medicinal plant of Polygonatum in Liliaceae,which is used as medicine by rhizome.In order to explore the effect of plant growth regulators on the growth of Polygonatum multiflorum rhizome sprouting seedlings,polygonatum multiflorum rhizome with lateral buds was used as experimental material.The rhizome of Polygonatum multiflorum was soaked in the mixture of KT,GA3and TDZ for 24 hours In order to provide theoretical basis for the application of kinetin(KT),gibberellin(GA3)and thidiazuron(TDZ)in rhizome seedling of Polygonatum multiflorum,the dynamic monitoring of endogenous hormone content,nutrient content and enzyme activity related to rhizome germination and seedling growth were carried out.The main results are as follows:1.After soaking Polygonatum multiflorum rhizomes with different concentrations of KT,GA3and TDZ mixture,the rhizomes of Polygonatum multiflorum all sprouted,while CK rhizomes of the control group did not sprout,and there were significant differences in rhizome emergence rate,survival rate,seedling height,seedling diameter and number of new buds among the treatments;treatment 5had the highest rootstock emergence rate(91.33%),treatment 1 the lowest(25.11%),treatment 6 had the highest survival rate(82.57%),and treatment 2 the lowest The results showed that the number of new shoots in treatment 9 was the highest(129.06cm),the number of new shoots in treatment 3 was the thickest(16.28mm),the number of new shoots in treatment 8 was the highest(2.24 per block),the number of new shoots in treatment 1 was the lowest(25.11%,30.00cm,6.98mm,0.21 per block).The results of orthogonal analysis of variance showed that KT,GA3and TDZ had different effects on rhizome emergence and growth of Polygonatum multiflorum.The effects on seedling emergence rate,seedling survival rate,seedling height,seedling diameter and number of new shoots were as follows:KT>GA3>TDZ,TDZ>GA3>KT,KT>TDZ>GA3,TDZ>GA3>KT,GA3>KT>TDZ.Duncan's test results showed that there were significant differences in seedling emergence rate,seedling height,number of new shoots in rhizome among KT levels,but no significant differences in seedling survival rate and seedling diameter;there were significant differences in seedling emergence rate,seedling height,seedling diameter,number of new shoots in rhizome among GA3levels,but no significant differences in seedling survival rate;there were significant differences in seedling survival rate,seedling height,seedling diameter,but no significant differences in seedling emergence rate and number of new shoots in rhizome among TDZ levels.There was a significant positive correlation between KT and seedling height,TDZ and seedling diameter,emergence rate and seedling height.The comprehensive evaluation of five agronomic traits of Polygonatum multiflorum showed that the comprehensive score of treatment5 was the highest,that is,the treatment of 200mg/LKT+100mg/L l GA3+600mg/LTDZhad the best effect on the growth of Polygonatum multiflorum.2.After soaking Polygonatum multiflorum rhizome with plant growth regulators,the content of endogenous GA3in rhizome of treatment 1?9 showed a trend of increase decrease from the early stage of rhizome germination to the early stage of rhizome seedling leaf development,and the differences among the treatments were significant;The content of endogenous GA3in rhizome of Polygonatum multiflorum was the highest in the early stage of seedling emergence,sprouting seedling stage,seedling bolting stage,seedling leaf unfolding stage and seedling leaf completely unfolding stage,and the content was 13.50%in treatment 4,8,5,8 and 5,respectively?g/g?27.05?g/g?22.78?g/g?30.96?g/g?32.83?g/g,Treatment 3 and CK were the lowest,respectively3.81?g/g?6.02?g/g?5.80?g/g?6.77?g/g?3.70?g/g.Except for treatment 2,the content of IAA in rhizomes of all treatments increased and decreased from the early stage of rhizome germination to the stage of complete leaf development,and there were significant differences among the treatments;The upward downward upward downward trend.In conclusion,exogenous plant growth regulators can indirectly affect the germination and growth of Polygonatum multiflorum by regulating the content and ratio of endogenous hormones in rhizome.3.After soaking Polygonatum multiflorum rhizomes with plant growth regulators,the SOD activity of rhizomes of Polygonatum multiflorum treated with 1-9 showed a trend of increase decrease increase,and the difference was significant with CK;The cat activity of treatment 1,4,5 and CK showed a decreasing increasing decreasing trend,while that of treatment 2,3,6,7 and 9 showed an increasing decreasing trend,which was significantly different from that of CK,but there was no significant difference between treatment 1 and 9;POD activity in rhizomes of treatment 1-9increased first and then decreased,and there was significant difference with CK,but there was no significant difference between treatment 1-9;The antioxidant enzymes SOD,cat and pod of rhizome maintained high activity from pre emergence to leaf development stage.The high activity antioxidant enzyme system was conducive to scavenging oxygen free radicals produced by rhizome sprouting seedlings and providing good conditions for their germination and emergence.The activity of MDH decreased,increased and decreased,while the activity of G-6-PDH increased first and then decreased,that is,TCA was transformed into PPP,and the proportion of PPP increased,which was beneficial to release dormancy of rhizome buds and promote germination of rhizome buds;Treatment 1?9 rhizomes?-Amylase and its application?-The amylase activity showed a trend of up down up,and the differences among treatments were significant?-Amylase and its application?-Amylase activity was significantly different in treatment 1?9?-Amylase and its application?-Amylase activity was higher than that of CK,indicating that plant growth regulators were beneficial to the growth of plants?-Amylase and its application?-The increase of amylase activity.4.After soaking with plant growth regulator,the soluble sugar content of rhizome1-9 decreased after the treatment,and the difference was significant among the treatments;At the seedling stage of rhizome sprouting,the soluble sugar content of1-9 rhizome reached the maximum,the highest was 25.72mg/g in treatment 1,and15.30mg/g in treatment 6;The soluble protein content of 1?9 rhizome showed a trend of rising decreasing rising,and the difference was significant with CK;The starch content of 1?9 roots decreased first and then increased,and the difference was significant with CK;The starch content of the first rhizome was at the lowest level and CK was at the highest level in the growth stage of Polygonatum rhizome.It was suggested that plant growth regulators could regulate the transformation between soluble nutrients and difficult nutrient nutrients in rhizome of Polygonum,and provide the necessary nutrients for the emergence of rhizome.
