Dinotefuran is the third-generation neonicotinoid insecticide newly developed after Imidacloprid and Thiacloprid.In recent years,neonicotinoid insecticides have attracted much attention due to their excellent properties.Folsomia candida is a soil model organism and arthropod,which can promote soil nutrient cycling and is of great significance to the ecosystem.However,with the widespread use of neonicotinoid insecticides,when studying the toxic effects of the first two generations of neonicotinoid insecticides on organisms,some scholars proposed that neonicotinoid drugs would affect the density of biological populations.In this study,the contact toxicity method was used to examine the toxic effects of dinotefuran on Folsomia candida.By measuring the activities of acetylcholinesterase,carboxylesterase,trace malondialdehyde and glutathione-s-transferase,the specific toxic mechanism of dinotefuran to Folsomia candida was understood.And through transcriptome sequencing and bioinformatics analysis,the specific toxicity mechanism was drawn and the following conclusions were drawn:For toxic effects.The results of the study showed that the acute toxicity LC50=0.75 mg/kg.A total of three concentrations of chronic toxicity were set at 0.002,0.006,and 0.018 mg/kg.The results of the study were that compared with the blank group in the concentration treatment group,the reproductive rate was reduced by 25%,29%,and 62%,respectively.The enzyme activities of glutathione-s-transferase,acetylcholinesterase,carboxylesterase and malondialdehyde were determined after exposure.The measurement results showed that acetylcholinesterase showed a downward trend as a whole,glutathione-S-transferase increased in the early stage,and the enzyme activity showed a downward trend with the increase of time,and the overall MDA activity showed an upward trend after exposure.Carboxylesterase showed no significant change in low concentration exposure in a short period of time,while the enzyme activity decreased at high concentration.Transcriptome analysis results showed that the differential genes were mainly concentrated in Baifutiao's nervous system,detoxification function and metabolic system,among which the effect on nerves was the most significant.The decrease of detoxification function and the decrease of metabolic function are the indirect causes of death of Folsomia candida.Finally,through the combined analysis of transcriptome analysis and enzyme activity assay results,the results of the two parts are consistent,and verified by q RT-PCR,the results obtained are consistent with the previous research results,and the experimental data are reliable.This study explores whether the third-generation drug dinotefuran has toxic effects and specific toxic effects from four levels,so as to understand its environmental fate and supplement toxicity data for the toxicity database.
