| Anxiety is one of common psychiatric disorders.There are usually awareness in advance,but inevitable and vicious incident to trigger an expected response,such as fear,chest tightness,anxiety,nervous and sleep disorder as the main performance,and often accompany with heart palpitations,sweat,respiratory distress and plant nerve dysfunction.So it has become an urgent problem that treating anxiety effectively.Albizzia bark is the dry bark of Albizzia julibrissin Durazz.It is often used for treating distraught,melancholy and ahynosis.There are so many researches in pharmacology of Albizzia julibrissin,but there are no literature research in(-)-syringaresnol-4-O-β-D-furan Qin glycosyl-(1→2)-β-D-glucopyranoside(for short,SAG),the index component of Albizzia julibrissin.It is necessary to study deeply on SAG,in order to elucidate the anxiolytic effect of mechanism.Therefore,based on early studies of Compound Valeriana jatamansi Jones,this research is intented to exoplre the anti-anxiety effect and mechanism of SAG in Albizia julibrissin bark,in order to expand its clinical application,to understand the effect of resolving stagnation for tranquilization,to establish its quality standards.It has theoretical and practical significance.This research is based on previous studies of anti-anxiety material basis and mechanisms,evaluating the anti-anxiety effect of SAG by using pharmacological models,and research on its anxiolytic effect mechanisms,also research preliminary on the anxiolytic effect of(-)-syringaresnol(for short,SYR),the aglycone of SAG,through animal experiments.The anxiolytic effect of SAG has been evaluated by pharmacological models and the mechanisms of its anxiolytic activity has been also explored.The main research contents and archieved results are as follow:1 The Pharmacological Research1.1 The anti-anxiety effect and mechanism of SAG on miceAccording to the results,in the doses of 6mg/kg/d,SAG could significantly increase OE%and OT%of mice in EPM.SAG of 6mg/kg/d could significantly increase time spent and transitions between light and dark compartments in LDB,times of central entries in the open-field.SAG of 3mg/kg/d exhibit anxiolytic effects in this three models,but it is not stable.However,these effects could be blocked by the antagonists Flu or WAY-100635.This indicated that the anxiolytic effect of SAG was block by WAY-100635 completely.In addition,SAG had no side-effect of activity in mice.1.2 The anti-anxiety effect of SAG and SYR on miceAccording to the research results,SAG of 3 mg/kg/d and SYR of 1.8mg/kg/d exhibit anxiolytic effects in EPM and LDB.From the results,Both SAG and SYR have certain anxiolytic effect,under the same circumstance,the effective dose of SYR is smaller than SAG Under the same condition,the anxiolytic effect of SYR is much better than SAG,but the relationship between SAG and SYR need further study.1.3 The anti-anxiety effect of SAG on rats with actue stressThe results show that,rats exhibit anxiety in EPM after the restraint of stress.In the doses of 3.6mg/kg/d,SAG could significantly increase the open entries percent and the open time percent of rats in EPM.2 Mechanism Studies2.1 Effects of SAG on monoamine neurotransmitter by stress exposureThe level of monoamine neurotransmitter(NE、5-HT and DA)and their metabolins(5-HIAA,DOPAC and HVA)in cerebral cortex and hippocampus of mice were analyzed by the HPLC-ECD.In cerebral cortex and hippocampus of mice,the concentration of NE,5-HT and 5-HIAA were declined after treatment of SAG,and also the level of DA and its metabol-ins in cerebral cortex of mice were declined.2.2 Effects of SAG on hypothalamic-pituitary-adrenal axis by stress exposureThe level of serum hormones in hypothalamic-pituitary-adrenal axis(for short,HPA axis)of the rats were measured by radioimmunoassay method.According to the results,the level of CRF,ACTH,CORT and GC were increased in acute stress rats which were negative control group.Compare with it,the doses of 3.6mg/kg/d,SAG could significantly decreased the level of CRF,ACTH,CORT and GC.The result indicated that 3.6mg/kg/d SAG had a stable and significant anxiolytic effect,and it might be related to HPA axis.3 ConclusionThe pharmacological models were used to evaluate the anxiolytic effect of SAG SAG is one of the principal component,also as anxiolytic effective component,in Albizzia julibrissin Durazz.The studies suggested that the anxiolytic effect of SAG might mediated by decreasing synthesis and release of monoamine neurotransmitters,and regulating HPA axis.Through the research,to establish a theoretical and experimental basis for the quality standard of Albizzia julibrissin Durazz. |
