Background Lung cancer is currently one of the leading causes of cancer deaths in the world.It is a malignant disease caused by various factors such as environment,genetics,and immunity.In the past half century,the incidence and mortality of lung cancer in various countries around the world have shown an increasing trend.Moreover,most of the currently diagnosed lung cancer patients are in the middle and advanced stages of lung cancer,and their five-year survival rate is extremely low.In contrast,the five-year survival rate of early-stage lung cancer presents a good prognosis,with a higher five-year survival rate.At present,the medical community urgently needs to find a safe and effective technical method that can assist clinical screening of early lung cancer.Objective Due to the rising incidence of lung cancer in China,and the limited early diagnostic methods leading to high mortality,we carried out A panel of autoantibodies(AABs),including CAGE,GAGE7,GBU4-5,MAGEA1,P53,PGP9.5,and SOX2,related to lung cancer in different populations Detection,and found its clinical application value for patients with early lung cancer,aiming to provide a reliable basis for early laboratory diagnosis of lung cancer patients.has been studied in the clinical setting to aid in the early diagnosis of lung cancer.Methods Here we report a study undertaken at Peking Normal Hospital in China to further evaluate the clinical performance of tHIS diagnostic kit.This study included serum samples of 121 newly diagnosed lung cancer patients,34 healthy subjects,50 patients with benign lung diseases and 100 human subjects with indeterminate solid nodules from the NationalComprehensive Cancer Network Lung Cancer Screening Guidelines.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of 7 autoantibodies in each group,and the ROC curve was drawn to analyze the diagnostic efficacy of lung cancer.The combined detection of 7 autoantibodies was positive among lung cancer patients with different clinical pathological features and between the groups.The rate and the positive predictive value of early lung cancer were observed by the results of 100 pulmonaryNodulespatients(staged follow-up of patients with undiagnosed suspected lung cancer).Results 1、Compared with healthy people,the concentration index of CAGE,GAGE7,GBU4-5,MAGE A1,P53,PGP9.5 and SOX2 in lung cancer group patients was greatly higher than that in the normal people control(P<0.05);with at least one of the seven antibodies being strongly positive as a criterion,the preliminary results showed a sensitivity of 45.5%(38%-56%,95%CI),specificity of 85.3%(73-95%,95%CI);the concentration index of seven antibodies in the lung cancer group patients was greatly higher than that in the non-lung cancer group control by the lung cancer group and the healthy group and the benign lung disease group(P<0.05);2、We made two comparisons for seven kit determination methods for lung cancer:①with at least one of the seven antibodies being strongly positive as the criterion for judging,the sensitivity was initially determined to be 45.5%(38%-56%,95%CI),specific The trait was 83.3%(75-94%,95%CI),According to the ROC curve,the AUCR was 0.660.The sensitivity and AUC were higher than those of the original universal tumor marker kit(name or type?).Specificity.The test result was determined to be positive if there was at least one strong positive antibody responses.The sensitivity was 45.5%(38%-56%,95%CI),with specificity at 83.3%(75-94%,95%CI),and AUC of ROC analysis was 0.660.②When the test result was determined to be positive if there was at least one strong positive antibody responses or at least two medium strong positive antibody responses,the sensitivity was increased to 55.4%while the specificity remained the same as 85.3%.The AUC of ROC curve was 0.719.③、The 7-AAb ELISA assay was also performed in 100 patients presented with GGNs or solid nodules of lung without diagnosis of any lung diseases.And the 6-month follow-up of the lung nodule group has been determined to predict the positive predictive value of lung cancer,he goal of the study was to follow up patients with positive test results to look for new incidence of lung cancer in prospective fashion.When 8 subjects were diagnosed with lung cancer and 2 subjects were diagnosed with benign lung diseases among 28 patients with positive test results.And information of the rest 18 subjects remained to be the same.The positive predictive value of the experiment was 72.7%.Conclusion So we report here that positive results of 7-AAb kit point to high risk of lung cancer.And 7-AAb panel showed high positive predictive values(PPV)to predict risk of lung cancer in patients with solid nodules.Our seven lung cancer autoantibodies types canprovide an important early warning sign in the clinical setting.
