Objective:To observe the effects of electroacupuncture on cerebral artery occlusion(MCAO)model of rats with neurological function deficit,cerebral infarction volume and expression of NR1\NR2A\NR2B in infarct brain tissue,so as to explore the mechanism of electroacupuncture promoting nerve repair.This study provides a theoretical and experimental basis for the clinical application of electroacupuncture at pericardial acupoints in the treatment of ischemic stroke(IS).Methods:A total of 180 SD rats were completely randomly divided into normal group(60 rats),and 120 rats needed to be modeled.MCAO rat model was prepared by modified thread embolization method.After successful modeling,MCAO rats were completely randomly divided into model group(60 rats)and electroacupuncture group(60 rats).Finally,the rats in the three groups were randomly divided into five subgroups with 12 rats in each subgroup at time points(1,3,7,14,and 21 days).The intervention was started24 hours after modeling.The electroacupuncture group took Tianquan,Quze,Neiguan and Daling acupoints and connected them to the electroacupuncture instrument for 30 minutes.The normal group and model group were held and bound the rats for 30 minutes,once a day,under the same conditions as the electroacupuncture group,and the corresponding subgroup was treated continuously for days.Before the first operation intervention and after the last intervention,the rats were scored according to Sun Jingfang's behavioral scoring standard.,executed after days of intervention corresponding subgroups based,the use of 2,3,5 triphenyl four azole nitrogen chloride(2,3,5-Triphenytetrazoliumchloride,TTC)dyeing,determination of cerebral infarction lesions volume,Western blot(WB)was used to detect the protein levels of NR1/NR2A/NR2 B in cerebral infarction tissues.Results:1.Behavioral score:(1)Compared with the normal group before treatment,there was statistical significance in the electroacupuncture group and model group(p<0.05);Compared with the model group,there was no statistical significance in each time point of the electroacupuncture group(p<0.05).(2)At the same time point,comparison before and after treatment:there was no statistical significance in the normal group(p>0.05);In the model group,the scores of the subgroup were decreased after 21 days(p<0.05);The scores of electroacupuncture subgroups were significantly decreased after 3d,7d,14 d and 21d(p<0.05).(3)At the same time point after treatment,compared with the model group,the subgroup scores of electroacupuncture group were decreased at 3,7,14 and 21 days after treatment(p<0.05).The results indicated that MCAO rats had a certain self-repair ability,and electroacupuncture at pericardial meridians was significantly better than that in model group in improving the behavior disorder of rats.2.Cerebral infarction volume: at the same time point,(1)compared with normal group,model group and electroacupuncture group were higher than normal group in 1d,3d,7d,14 d and 21d(p<0.05),indicating that the model establishment was successful.Compared with model group,electroacupuncture group had no significant difference in 1d and 3d(p>0.05),but decreased in 7d,14 d and 21d(p<0.05),indicating that electroacupuncture could promote the reduction of cerebral infarction volume.3.NR1/NR2A/NR2 B protein content:(1)Protein content of NR1:At the same time point,(1)the expression of NR1 in model group increased at 1d,3d,7d,14 d and 21d(p<0.05),while in electro-acupuncture group,the expression of NR1 in 1d,7d,14 d and 21d(p<0.05)increased,but the difference was not significant(p>0.05).Compared with the model group,the expression of EA group in 1d,3d,7d,14 d and 21 d was lower than that in the model group(p<0.05).The results showed that the expression of NR1 was promoted after cerebral ischemia in rats,and electroacupuncture could inhibit the increase of NR1.(2))Protein content of NR2A:At the same time point.(1)Compared with the normal group,the content of NR2 A protein in the model group was increased at 3d(p<0.05),and decreased at 14 d and 21d(p<0.05).In the electroacupuncture group,it decreased on day 1,3 and 7(p<0.05),and there was no significant difference on day 14(p<0.05),but it increased on day 21(p<0.05).(2)Compared with the model group,the electroacupuncture group decreased on day 1,3 and 7(p<0.05);Increased on 14 and 21d(p<0.05).The results showed that the expression of NR2 A in ischemic brain tissue of MCAO rats increased firstly and then decreased.Electroacupuncture intervention in the early cerebral ischemia can reduce the content of NR2 A protein.The protein content was increased in the middle and late stage.(3)NR2B protein content:At the same time point.(1)Compared with the normal group,it was increased on day 1,3 and 7 in model group(p<0.05),and decreased on day 14 and 21(p<0.05).There was no significant difference in electroacupuncture group on day 1(p<0.05),increased on day 3(p<0.05),decreased on day 7(p<0.05),and increased on day 14 and 21(p<0.05).(2)Compared with model group,electroacupuncture group was lower than model group on 1d,3d and 7d(p<0.05),and higher than model group on 14 d and21d(p<0.05).The results showed that the expression of NR2 B in ischemic brain tissue of MCAO rats increased firstly and then decreased.Electroacupuncture can inhibit the expression of NR2 B in the early stage of cerebral ischemia and promote its expression in the middle and late stage.Conclusion:1.Electroacupuncture at Tianquan,Quze,Neiguan and Daling points can improve the motor function and reduce the volume of cerebral infarction in MCAO rats.2.Electroacupuncture can improve the motor function and reduce the volume of cerebral infarction in MCAO rats by inhibiting the expression of NR1/NR2A/NR2 B at the early stage of cerebral ischemia,inhibiting the expression of NR1 and promoting the expression of NR2A/NR2 B at the middle and late stage of cerebral ischemia,thus activating the survival pathway of nerve cells,repairing damaged nerve cells and improving nerve function.
