Objective:To observe the effect of electroacupuncture at pericardium meridian acupoint on neurological deficit score,serum Nogo-A and expression of Nogo-A and NgR1 in cerebral infarction area of MCAO rats,and to explore the possible mechanism of electroacupuncture at pericardium meridian acupoint to promote nerve repair from Nogo-A/NgR1 pathway,so as to provide experimental basis and theoretical support for clinical application of pericardium me-ridian of hand in the treatment of cerebral infarction.Methods: The 150 rats were randomly divided into normal group(n=30),sham operation group(n=30)and model group(n=90).After modeling,90 model rats were randomly divided into model group(n=30),pericardium meridian acupoint group(n=30)and lung meridian acupoint group(n=30),and then were randomly divided into 3d,7d,14 d,21d,28 d 5time points,with 6 rats in each time point.MCAO model rats were made by improved thread occlusion method,and the acupoints of different segments of pericardium meridian and lung meridian were selected for electroacupuncture treatment on the second day after successful modeling.The neurological deficit score was observed,and the content of serum Nogo-A was detected by ELISA,and the expression of Nogo-An and NgR1 in ischemic brain tissue was detected by immunohistochemistry and RT-PCR method.Result:1.Neurological deficit score of MCAO rats:With the passage of time after cerebral infarction,the neurological deficit scores of rats in pericardium meridian group,lung meridian group and model group decreased in varying degrees.Compared with 12 h,there was significant difference in model group in 21 d and 28d(p<0.05,p<0.01).The score decreased significantly after 14 d in pericardium meridian group and lung meridian group(p<0.05,p<0.01).Compared with normal group and sham operation group,the scores of pericardium meridian group,lung meridian group and model group were higher at each time point(p<0.01),and the pericardium meridian group and lung meridian group could lower the neurological function score in different degrees,but the difference was not statistically significant compared with the model group(p>0.05).2.The Nogo-A in Serum: Intra-group comparison: In the model group,the expression of Nogo-A in the model group decreased in 7d and 28 d compared with that in 3d,but there was no significant difference at each time point in the lung meridian group,except that in the lung meridian group,the expression of Nogo-A 3d in the pericardium meridian group increased gradually,reached the peak in 14 d,and decreased continuously in 21 d and 28d(p<0.01,p<0.05).Group comparison: Compared with the normal group and the sham operation group,the expression of Nogo-A in the pericardium meridian group,the lung meridian group and the model group increased at each time point,and the expression in the pericardium meridian group was lower than that in the model group in 3d,21 d and 28d(p<0.01,p<0.05),and it was lower than that in the lung meridian group in 3d and 21d(p<0.01).3.The Nogo-A in Cerebral ischemic area: Intra-group comparison: The expression of Nogo-A and Nogo-A m RNA in normal group changed little at each time point in sham operation group(p>0.05),while the expression of Nogo-A and Nogo-A m RNA in model group showed a certain regularity,showing a fluctuating upward trend(p<0.01)Lung meridian group: Nogo-A: 21d>7d,28d> 3d and 14 d,Nogo-A m RNA: 28 d,21d>14d>3d and 7d(p<0.01,p<0.05);In pericardium meridian group: nogo-a: 21d>7d>28d>14d>3d,Nogo-A m RNA: 28 d,14d>21d,7d and 3d(p<0.01,p<0.05).Group comparison: Model group: Nogo-A and Nogo-A m RNA were higher than those of normal group and sham operation group at each time point(p<0.01).The expression of Nogo-A in pericardium meridian group and lung meridian group was lower than that in model group(p<0.01).And the NgR1 in the pericardium meridian group was lower than that in the lung meridian group on the 21 d and 28d(p<0.01),while the NgR1 m RNA in the pericardium meridian group was lower than that in the model group at all time points(p<0.01),and it was lower than that in the lung meridian group except 14 days(p<0.01),and the expression of NgR1 m RNA in the lung meridian group was lower than that in the model group on the 3d and7 d.4.The NgR1 in Cerebral ischemic area: Intra-group comparison: In sham operation group,NgR1 and NgR1 m RNA did not change significantly at each time point.In the model group,NgR1:28d,14d>21d,7d>3d,NgR1 m RNA:28d,3d>21d>7d,14d(p<0.01),lung meridian group: NgR1:28d,21d>14d>7d>3d,7d(p<0.01),pericardium meridian group:NgR1:28d>21d>3d,7d(p<0.01,p< 0.05),the lung meridian group: NgR1:28d,21d>7d>3d,7d(p<0.01,p<0.01),The lung meridian group: NgR1 m RNA:28d>14d>21d>3d,7d(p<0.01).Group comparison: NgR1 and NgR1 m RNA in model group were higher than those in normal group and sham operation group,the expression of NgR1 in lung meridian group and pericardium meridian group was lower th-an that in model group at all time points(p<0.01),and lower than that in lung meridian group at 21 d and 28d(p<0.01).The NgR1 m RNA of the lung meridian group was lower than that of the model group on the 3d and 7d,but higher than thatof the model group on the 14 th day,and lo-wer than that of the model gro-up at each time point in the pericardium meridian group(p<0.01).Except for 14 d,it was lower than that of the lu-ng meridian group(p<0.01,p<0.05).Conclusion:1.After MCAO modeling,the expressions of Nogo-A and NgR1 in the cerebral tissue of the infarct side of rats showed A fluctuating upward trend at each time point.2.Electroacupuncture of pericardium meridian and lung meridian can accelerate the recovery of neurological function in MCAO rats and Electro-acupuncture of pericardium meridian can inhibit and stabilize the expression of Nogo-A in serum and Nogo-A and NgR1 in infarcted brain tissue.3.Electroacupuncture at pericardium meridian points may accelerate the recovery of neurological function in MCAO rats by down-regulating the expression of Nogo-A and NgR1.
