Objective: The effects of Fucong tablet on the content of myelin basic protein(MBP)and the structure of myelin sheath in the cochlear nucleus of C57BL/6J mice were observed by auditory brainstem response(ABR),transmission electron microscopy(TEM)and Western blot,Objective to explore the mechanism of Fucong tablet in the prevention and treatment of presbycusis.Methods: Fifty six 4-week-old C57 BL / 6J mice of clean grade,half male and half female,were fed adaptively for one week.14 mice were randomly selected as young blank group.After ABR detection,the mice were decapitated and the cochlear nucleus was frozen in liquid nitrogen.The remaining 42 mice were fed for 5 months and randomly divided into three groups after ABR detection.Group A was treated with Fucong tablets(fed with Fucong tablets),group B was treated with Ginkgo biloba capsules(fed with Ginkgo biloba umbilical capsules),and group C was the elderly blank control group(fed with normal feed).The ABR was detected again after 3months.ABR thresholds of the three groups were compared before and after treatment,the myelin sheath in cochlear nucleus was observed by transmission electron microscope,and the content of MBP in cochlear nucleus was detected by Western blot.Results: 1 ? The difference of hearing threshold before and after treatment in Fucong tablet group,Ginkgo biloba group and elderly blank control group was 16.54 dbspl,19.17 dbspl and 22.50 dbspl,respectively.There was significant difference between Fucong tablet group and elderly blank control group(P < 0.05),but there was no significant difference between Ginkgo biloba capsule group and elderly blank control group(P > 0.05);2? In the old blank group,the diameter of axons became larger,the arrangement of myelin cells was disordered,the structure of myelin cells was fuzzy,the lamellar structure was loose and broken;there were large mitochondria in axons and lipofuscin deposition in cytoplasm;in the Fucong tablet group,the myelin structure was slightly clear,the myelin cells were slightly neat,and the axon swelling was slightly weakened compared with the old blank group;3? Compared with the young blank group,the content of MBP in the cochlear nucleus of the old blank group was decreased,the difference was statistically significant(P < 0.05);compared with the old blank group,the expression of MBP in the Fucong tablet group and the Ginkgo biloba capsule group was increased(the Fucong tablet group was higher than the Ginkgo biloba group),but the difference was not statistically significant(P > 0.05).Conclusion: Fucong tablet can effectively delay hearing loss in aged C57 BL /6J mice,but whether its mechanism is related to MBP content in cochlear nucleus and myelin sheath structure remains to be further proved.
