Objective:Sort out and analyze the prescriptions and medication rules of traditional Chinese medicine for the treatment of Helicobacter pylori-related gastropathies,and explore its effective ingredients and mechanism of action.Methods:1.Search the Chinese journal full-text database,Wanfang data knowledge service platform,Weipu.com,and China Biomedical Literature Database for literature on the treatment of HP-related gastric diseases with integrated Chinese and Western medicine as data sources,screen and use Excel software and TCM inheritance auxiliary platform to establish a prescription database Perform statistical analysis and excavate drug usage rules,and analyze core drugs.2.Forty SPF BALB/c mice were randomly divided into normal group,model group,drug pair group(pinellia-Scutellaria-Coptis),standard quadruple group,a total of 4 groups,each with 10 mice.Except for the normal group,the other three groups were given intragastric administration of Helicobacter pylori bacteria liquid to establish a mouse model of Helicobacter pylori-associated gastritis.After the model was successfully established,except for the normal group and the model group,the mice in the other groups were given distilled water gavage.The corresponding drugs were given by gavage for 14 days.Rapid urease experiment and modified W-S silver staining method to detect Helicobacter pylori infection and colonization;HE staining method to detect the pathological changes of gastric mucosal tissue;ELISA method to detect serum IL-1? and TNF-? levels in each group of mice.3.Screen the potential active ingredients of the"Panxia-Scutellaria-Coptis" drug pair through the TCMSP platform;the Swiss Target Prediction database to obtain the target of potential active ingredients,and the Gene Cards database and OMIM database to obtain the main pathogenic targets of HP-related gastritis Point;construct a "Chinese medicine-component-target" network model and PPI network diagram to analyze the main active ingredients and target proteins in the drug pair;use the David database to analyze the biological function and metabolic pathways of the drug pair target.Results:1.Literature analysis included a total of 178 qualified literatures,77 of which clearly defined TCM syndromes,with the highest frequency of spleen and stomach damp-heat syndrome;a total of 207 prescriptions were used,of which the classic prescription was used 100 times,and Banxia Xiexin Decoction was used the most frequently;There were 3,027 Chinese medicines,including 207 different Chinese medicines.The top three single Chinese medicines used were licorice,coptis and pinellia.The four qi of traditional Chinese medicine are both cold and warm,and the five flavors are mostly bitter and pungent,mostly attributable to the spleen meridian,stomach meridian and liver meridian.Data mining technology was used to mine 21 core drug combinations and 12 new prescriptions were obtained.2.Rapid urease and modified WS silver staining method indicated successful Helicobacter pylori infection;2 weeks after administration,the rapid urease test results indicated that the eradication rate of HP in the standard quadruple group was 8 8.89%,and the eradication rate of HP in the drug-pair group was 77.78%,There was no statistical difference between the two groups(P>0.05);the HE staining results of gastric tissue showed that the gastric mucosal structure of the model group was partially damaged,and the inflammatory cell infiltration was obvious,while the standard quadruple group and the drug caused less damage to the gastric tissue structure.There was a little inflammatory cell infiltration;Elisa results showed that compared with the normal group,the serum levels of IL-1? and TNF-? in the model group were significantly increased,and the difference was statistically significant(P<0.01).After the intervention of standard quadruple drugs and drug pairs,the levels of IL-1? and TNF-? decreased,and compared with the model group,the difference was statistically significant(P<0.05).There was no difference between the standard quadruple group and the drug pair group.3.35 potential active ingredients of the drug were screened out,among which compounds with higher Degree values include baicalein,baicalein,nor wogonin,etc.;37 potential target proteins of the drug for the treatment of HP-related gastritis It mainly includes EGFR,STAT3,MAPK,HSP90,MMP,MTOR,NOS3,etc.;77 pathways are involved,including cancer pathway,HIF-1 signaling pathway,TNF signaling pathway,FoxO signaling pathway,cholinergic antiinflammatory pathway,etc.Conclusion:1.HP-related gastropathy is located in the spleen and stomach,which is related to the liver and lungs;commonly used heat?clearing drugs,tonic drugs and qi regulating drugs,the drugs are used in combination with cold and temperature,the medicine tastes bitter and sweet,and the treatment methods are mainly from"exorcise evil"and "tonic It is in accordance with the pathogenesis of the deficiency and excess,and the mixed cold and heat.The medication has the characteristics of the combination of cold and temperature,and pungent to relieve bitterness.Among them,the combination of pungent prescribed and bitter-reducing drugs to "pinellia-CoptisScutellariae" may be the key drug for the treatment of HP-related gastropathies.2."Banxia-Coptis-Scutellaria" has the effect of antiH.pylori and alleviating gastric mucosal inflammation.3.PinelliaCoptis-Scutellaria baicalensis" intervention HAG has the characteristics of multi-component,multi-target,multi-channel treatment,which provides a theoretical basis for further experimental research on the material basis and mechanism of its pharmacodynamics.
