Objective:1.Clinical partThrough the analysis of the clinical medical records of Professor Yao-sheng Zhang’s treatment of oligoasthenospermia,in-depth study of Professor Yao-sheng Zhang’s experience in treating oligoasthenospermia.Analyze the law of addition and subtraction and combination of drugs and explore modern statistical research methods of passing on experience in traditional Chinese medicine(TCM).2.Network pharmacology partAccording to the clinical research results,guided by the basic theories of TCM,the research methods of network pharmacology are used to explore the possible mechanism of core drugs in the treatment of oligoasthenospermia,providing a preliminary basis for in-depth experimental research.Methods:1.Clinical partEstablish a TCM clinical prescription database for Professor Yao-sheng Zhang to treat oligoasthenozoospermia,using the "Traditional Chinese Medicine Inheritance Computing Platform V(3.0)" software provided by China Academy of Chinese Medical Sciences,and analyze prescriptions using frequency statistics,association rules,and mutual information methods The rule of medication.2.Network pharmacology partSelect the effective elements and targets of core drugs by searching the Traditional Chinese Medicine System Pharmacology Platform(TCMSP);select the disease targets of oligoasthenospermia by searching the human gene information database;screen the common targets of drugs and diseases,and use Cytoscape software to construct Drug-disease-target interaction network;construct a common target protein interaction network(PPI)through the STRING platform;use R language to perform GO and KEGG enrichment analysis on the common target of prescription core drugs and oligoasthenospermia.Results:1.Clinical partThis study screened 163 medical records of oligoasthenozoospermia,with a total of 213 prescriptions.The age composition of the patients is between 26 and 50 years old;107 kinds of symptoms are involved;47 kinds of tongue images and 51 kinds of pulse conditions;drug statistics involve 152 kinds of Chinese herbal medicines,and the total frequency of drug use is 3568 times.There are the most types,the main medicine properties are flat,warm,and cold medicines,and the main medicine tastes are sweet and spicy.Based on the association rule Apriori algorithm,the core drug combination is extracted and the algorithm is verified.2.Network pharmacology partAfter preliminary screening,97 active ingredients of the four core drugs were obtained.Among the active ingredients of medicine,the highest degree value is quercetin,luteolin,β-sitosterol,glycitein,and stigmasterol.In genes,the first degrees of degree are NCOA2,PTGS2,PTGS1,AR,and CASP3.46 common targets of oligoasthenozoospermia diseases and drugs were retrieved,and the PPI network diagram and frequency diagram drawn by the R language were AKT,IL6,ESR1,EGFR,etc.with higher frequency.The enrichment analysis results of GO and KEGG pathways mainly include biological processes such as oxidation activity,phosphatase binding,and endogenous hormone regulation.Conclusions:1.Clinical partIn this study,medical records of Professor Yao-sheng Zhang’s clinical treatment were sorted through the development of a standard information statistical program,from the basic information,symptoms,tongue diagnosis,pulse diagnosis of the patient to the extraction of prescription drugs,and the extraction and algorithm of core drug combinations.Analyzed from multiple angles such as verification,the results obtained initially reveal the relationship between disease-symptoms-prescriptions and medicines of patients with oligoasthenospermia,and provide more ideas for the promotion and application of TCM in oligoasthenospermia.2.Network pharmacology partQuercetin,luteolin,β-sitosterol,glycitein,and stigmasterol may play a role in the pharmacological effects of Chinese herbal drugs for invigorating the kidney and spleen.The possible targets of Chinese herbal drugs for invigorating the kidney and spleen in the treatment of male oligoasthenospermia are AKT,IL6,ESR1,EGFR,etc.In addition,the role of ESR1 as an estrogen receptor gene in regulating male spermatogenesis is clearly reflected in this study.The study speculates that the mechanism of kidney-tonifying and spleen-invigorating Chinese medicine for improving male oligoasthenospermia and participating in the regulation related to endogenous hormones.
