[Objective]This project intends to explore the treatment method of silicone sheet combined with stretching force reduction,and to explore the precise quantitative evaluation of silicone sheet for skin stretching force reduction,so as to provide experimental research basis for clinicians to apply silicone products to prevent keloid.[Method]Selecte silicone sheet(Molnlycke Healthy Care,lnc.Production,approval number:National Machinery injection 20172640642,specification:10*18cm)at room temperature,and use high-precision tensile tester(China Dongguan Zhiqu Precision Instrument Co.,Ltd.,model 990L)to test the stretching force of the different specifications of the sheet stretching to the different deformation rate.According to the stretching force of different deformation rate,draw the corresponding relationship graph.[Results]The width of Mepiform was kept at 2cm,and the length started from 2cm,each length was increased by 0.5cm,and the longest length was 10cm for the experiment.After stretching to deformed the Mepiform,its resilience and viscosity could provide stretching force reduction for local skin.After the tester reached the setting deformation value,the force value continued to decay.The stretching force of the MEPIFORM tends to be stable when the force is well-distributed.Through experiments,the range of maximum force value is 1.44N-4.54N of Mepiform in different sizes the under 5%deformation,the range of stable force value is 0.97N-2.57N.The difference between maximum force value and the stable force value in the same size is 1.97N;the range of maximum force value is 4.19N-7.51N of Mepiform in different specifications the under 10%deformation,the range of stable force value is 1.99N-5.54N.The difference between maximum force value and the stable force value in the same size is 2.20N;the range of maximum force value is 7.74N-13.98N of Mepiform in different specifications the under 15%deformation,the range of stable force value is 3.87N-8.44N.The difference between maximum force value and the stable force value in the same size is 5.54N;the range of maximum force value is 12.71N-21.65N of Mepiform in different specifications the under 20%deformation,the range of stable force value is 6.92N-13.89N.The difference between maximum force value and the stable force value in the same size is 7.76N;draw the stretching force-deformation relationship graph of the silicone sheet under different sizes,and the area graph of the difference between the maximum force value and the stable force value of Mepiform under different level deformations.In 5%and 10%deformation,Mepiform can provide a few stretching force reduction,and at this time,the difference between the maximum force value and the stable force value is the smallest in each size of Mepiform,and effect of the stretching force reduction is more stable;In the deformation of 15%and 20%,Mepiform can provide larger stretching force reduction the maximum force value and the stable force value are quite different,and the stretching force reduction effect is unstable.[Conclusion]Different sizes of Mepiform can provide stable reduced tension value though 5%and 10%deformation of Mepiform.It is suitable for applicating in wounds with less tension.It is convenient for patient using silicone sheet by themselves to obtain stable and continuous compression value of tension.When the deformation keepin 15%and 20%,silicone sheet can provide greater value of stretching force reduction,and it is more suitable for those wounds with greater local tension after surgery.According to the tension graphs,clinicians might explore more suitable treatment methods in different sizes of skin lesions,and provide new ideas and experimental research basis for future scar prevention and treatment.[Objective]To explore the correlation between the expression of focal adhesion kinase(FAK)in different stretching force positions in keloids which occured on shoulders and nomal control.[Method]Three samples selected from patients with keloids occurred on any shoulder.Using immunohistochemistry(IHC)to observe FAK expression in central,edge of keloid and normal control.Each tissue in the edge group and the central group was made into 2 slices,and each slice was taken 5 visual fields,and each case in the normal group takes 5 visual fields.Use ImageJ image analysis software to analyze the integrated optical density(IOD)value of each field,use SPSS25.0 data analysis software to statistically analyze the experimental results.[Results]The quartile data of the IOD value of the edge tissue is 40704.54(37824.44,56196.95),the quartile data of the IOD value of the central tissue is 26030.40(16608.04,37283.15),and the quartile data of the IOD value of the normal control group is 1692.05(678.41,13712.69).Through rank sum test,the expression of FAK in the edge tissues of keloids was more than that of the central tissue(P<0.05),and the expression of FAK in the normal tissue was significantly decreased compared with the edge group and the central group(P<0.05),showing statistical difference.[Conclusion]1.FAK might be the biomarker for keloids;2.The expression of FAK in keloid tissue is proportional to skin tension,showing the growth pattern of keloid in different parts of human may be affected by local tension;3.FAK is expected to be used as a therapeutic target for keloids for further research.
