Objective To investigate the effects of gastrodin on the medial vestibular nucleus of rats with unilateral labyrinthine damage(UL)The effect of gastrodin on the expression of H1,H2,H3 subtypes of nucleus(MVN)histamine receptor was studied.The results of this study can reveal the possible mechanism of gastrodin promoting vestibular function compensation,and provide new ideas for the prevention and treatment of vestibular dysfunction and promoting vestibular function compensation in the future.Material and method 192 adult healthy SD rats were randomly divided into blank control group(n=12),sham operation group(n=36)and model group(n=144).The model rats were made by injecting 50%chloroform into the tympanum to duplicate the unilateral labyrinthine destruction model.The sham operation group was made by the same operation,but the tympanum was injected with the same amount of normal saline.After model evaluation,the model rats were randomly divided into four groups:model control group(n=36);high dose group(n=36);medium dose group(n=36);low dose group(n=36).Rats in the blank control group were fed normally without any treatment;rats in the sham operation group and the model control group were injected with normal saline(75mg·kg-1·D-1)once a day for three days;rats in the high,medium and low dose groups were injected with high(100mg·kg-1·D-1,middle(75mg·kg-1·D-1),low(50mg·kg-1·D-1)doses of gastrodin once a day for three days.After the last administration,the rats were killed.The expression of histamine receptor H1,H2,H3 m RNA and protein in the medial vestibular nucleus was observed by RT-PCR,immunohistochemistry and Western blot.SPSS 22.0 statistical software was used to analyze the experimental data.The measurement data were first tested for homogeneity of variance,and then one-way ANOVA was used for one-way ANOVA.LSD was used for the comparison between the two groups.The measurement data were expressed as mean±standard deviation,with P<0.05 as the statistical difference.Results On the 1st,3rd and 7th day,there was no significant difference in the expression of histamine receptor H1,H2,H3 m RNA and protein between the sham operation group and the blank control group(P>0.05);on the 1st and 3rd day,compared with the blank control group,there was no significant difference in the expression of histamine receptor H1,H2,H3in the model control group Compared with the model control group,the expression of histamine receptor H1,H2,H3 m RNA and protein in MVN of gastrodin high dose group was significantly up-regulated(P<0.05),But on the 7th day,there was no significant difference between the above groups(P>0.05).Conclusion The up-regulation of histamine receptor H1,H2,H3 m RNA and protein expression level in MVN may be one of the important mechanisms of vestibular compensation in rats;the up-regulation of histamine receptor H1,H2,H3 m RNA and protein expression levels in the medial vestibular nucleus of rats in each dose of gastrodin intervention group showed varying degrees,indicating that gastrodin can promote the expression of H1,H2,H3 receptor genes and proteins.The results showed that the high dose group was significantly better than the middle and low dose group,showing a dose-dependent relationship.
