Purpose:To investigate the effect of Dioscorea nigra on the expression of PDX-1mRNA and Fas protein in pancreatic tissues of type 1 diabetic mice,to further explain the anti-apoptotic effect and mechanism of dioscin on pancreatic islet ? cells in type 1 diabetic mice.It is extracted from the traditional Chinese medicine Dioscorea nigra Dioscin protects pancreatic islet ? cells and provides a theoretical basis for improving pancreatic islet function.Materials and methods: Eighty-nine SPF-grade male C57BL/6 mice,6-8 weeks old,were properly fed in an SPF-grade animal breeding room for 1 week.Twelve mice were randomly selected as the normal control group,and the remaining 77 mice were used for modeling.After 12 hours of fasting,the mice were injected with a well-prepared and cryopreserved STZ solution(PH=4.2)into the abdominal cavity of the mice.),the injection dose is 50mg/kg,and the injection is repeated for 5 consecutive days to induce the mouse to become a type 1 diabetes model.The blood glucose value in the tail vein of the mouse was measured 72 hours after the last intraperitoneal injection of STZ with a blood glucose meter.Those greater than 16.7mmol/L were considered as successful experimental modeling,and the successfully modeling mice were randomly divided into 4 groups,named model group,dioscin low-dose group,dioscin high-dose group,and prednisone group.Both the model group and the treatment group started gavage after the model was successfully established,and administered once a day until the end of the 12 th week of the experiment,during which the general status,blood sugar and weight of the mice were observed.PCR was used to detect the expression level of PDX-1mRNA in mouse pancreas,and WB technology was used to detect the expression of Fas protein in mouse pancreas.During the whole experiment period,all mice were fed with normal mouse growth and breeding feed,free movement,eating and drinking.Results:1.The mice in the model group have poor mental state,dull coat color,thin body,slow response,increased eating and drinking,and polyuria.The mice in the administration group also showed the above symptoms,but the symptoms were lighter than the model group,and their body weight was heavier than the model group.2.Compared with the normal control group,the blood glucose level of mice in the model group was significantly increased(P<0.01);from the 4th week of administration,the blood glucose of the diosgenin treatment group was significantly lower than that in the model group(P<0.01).0.01),the prednisone group was not statistically significant compared with the model group.3.4 weeks after modeling,compared with the normal control group,the body weight of the model group,dioscin low-dose group,dioscin high-dose group,and prednisone group decreased significantly(P<0.01);from 8 weeks From the beginning to 12 weeks later,the body weight of the mice in the high-dose treatment group increased to a certain extent compared with the model group(P<0.01).4.Compared with the normal control group,the expression of PDX-1mRNA in the model group was significantly reduced(P<0.01);compared with the model group,the expression of PDX-1mRNA in the pancreas of the mice in each treatment group was significantly increased(P<0.01)).5.Compared with the normal control group,the expression of Fas protein in the pancreas of the model group was significantly increased(P<0.01);compared with the model group,the expression of Fas protein in the pancreas of each treatment group decreased(P <0.01),the expression of Fas protein in the pancreas of mice in the dioscin high-dose group was significantly reduced(P<0.01).Conclusion:Dioscorea nigra saponins can improve the mental and behavioral status of type 1 diabetic mice,increase the weight of mice,reduce blood sugar,and at the same time increase the expression level of PDX-1mRNA in mouse pancreatic tissue,down-regulate the expression level of Fas protein,and inhibit pancreatic ? Apoptosis,protects pancreatic ?-cell function,and slows down the symptoms of diabetes.
