Purpose:In this study,DSS was used to prepare the rat model of ulcerative colitis.Bilateral Tianshu and Zusanli points were selected and electroacupuncture was used to intervene.At the same time,16 SrDNA high-volume sequencing technology was used to explore the protective effect of electroacupuncture on intestinal microecology and the key flora affected by electroacupuncture.To study the therapeutic effect of acupuncture and moxibustion on ulcerative colitis from the perspective of intestinal flora,and to provide a practical basis for in-depth discussion of the mechanism of acupuncture and moxibustion in the treatment of inflammatory bowel disease.It also provides a deep theoretical reference and experimental basis for the comprehensive understanding of the efficacy of acupuncture and moxibustion,which is a multi-target and multi-level complex system.Materials and Methods:Forty healthy male SD rats,aged 8 weeks and weighing300±15g,were fed adaptively for 3 days before the experiment began.SPSS26.0 software was used to randomly divide them into blank group,model group,electroacupuncture treatment group and non-meridian and non-acupoint treatment group,with 10 rats in each group.Rats in blank group were given distilled water for free intake,and the other 3 groups were treated with Dextran sodium sulfate(DSS)to prepare the immune response rat model of ulcerative colitis.In the electroacupuncture group,Tianshu and Zusanli acupoints on both sides of rats were selected for electroacupuncture treatment,while in the non-non-acupoint treatment group,bilateral Tianshu acupoints and the midpoint of the line of the fourth ipsilateral nipple and Zusanli acupoint were selected for electroacupuncture treatment,and the electroacupuncture method was the same as that of the electroacupuncture treatment group.The blank group and the model control group were only bound for the same time without electroacupuncture,10 min every time,once every 24 h,for consecutive 7 days.During this period,the model group,the electroacupuncture group and the non-acupoint group continued to freely drink sodium dextran sulfate(DSS),and the blank group freely ingestion of distilled water.After the intervention,disease activity(DAI)index was used to evaluate the severity of colitis.At the same time,fresh feces of four groups of rats after treatment were collected for fecal occlusion blood test,and 16 S rDNA ampland sequencing method was used to measure the intestinal flora.Finally,the rats were anesthetized by abdominal injection.After the rats lost consciousness,the abdomen was dissected,the abdominal aorta was intubated and the blood was extracted,and the tissues of the distal colon and ileocecal part were intercepted,and the pathological morphology of the rats was observed.SPSS 26.0 software was used for statistical analysis.Results:1.Influence of electroacupuncture intervention on UC rat model made by DSS,(1)DSS induced ulcerative colitis rats compared with the blank group,model group,the basic situation of the hair color dim and there is no luster,reaction force is weak,adhesion and waste around the anus,quality of a material is thin or stool watery stools,visible waste mixed blood or secretions purulent blood samples,activity intensity is abate,weight loss,DAI rating numerical significantly increased compared with the blank group(P < 0.05);Compared with the model group,the activity intensity of UC rats in the electroacupuncture group was increased,and the diet and drinking water were better.The body weight increased,the mental condition improved significantly,and the hair color recovered luster.Less sparse stool,no more blood in stool.DAI score was lower than that of model group,and the difference was statistically significant(P < 0.05).Compared with the model group,there were no significant differences in daily activities,diet and drinking water,body weight,mental state,hair color,fecal traits and blood in stool of UC rats in the non-meridian and non-acupoint group,and there was no significant difference in DAI score between the model group and the non-meridian and non-acupoint group(P > 0.05).(2)Compared with the blank group,the colonic tissue of rats in the model group showed different size of ulcer surface,hyperplasia of mucosa and submucosa,infiltration of a large number of neutrophils,and disordered arrangement of glandular structures.After electric acupuncture intervention,the curative group rats colon tissue morphology obviously improved,mucosal hyperemia situation improved,edema is mitigated,ulcer is also was restored,seen by the microscopic colonic tissue glands number increased,and glandular structures arranged orderly,inflammatory cells to reduce or disappear,mucosal epithelial tissue to restore the continuity;There was no significant difference between the non-meridian and non-acupoint group and the model group.2 Detection of intestinal flora16S rDNA amplicon sequencing showed that compared with the blank group,the number of OTUs,alpha diversity and beta diversity in the model group were decreased,and the abundance and diversity index of intestinal flora in the model group were significantly decreased(P < 0.05),which was statistically significant.Compared with the model group,there were significant differences in the number of OTUs,alpha diversity and beta diversity in the electroacupuncture group,and there were significant increases in the abundance and diversity index of intestinal flora in the electroacupuncture group(P < 0.05),which was statistically significant.The number of OTUs,the diversity of Alpha and the diversity of Beta in the non-meridian and non-acupoint groups were also different,but the number of OTUs in the non-acupuncture group was significantly different(P < 0.05).The dominant bacteria mainly belong to Proteobacteria,Bacteroidetes and Firmicttes.After electroacupuncture treatment,the number of the corresponding flora showed a trend to the normal direction compared with that before acupuncture treatment.Conclusion:1.Electroacupuncture at "Tianshu" and "Zusanli" can effectively reduce DAI score,increase body weight and improve intestinal histomorphological inflammatory manifestations of UC rats.2.Electrotherapy can positively regulate the diversity of intestinal microflora and the content of beneficial bacteria in UC model rats.3.Electroacupuncture has a protective effect on intestinal mucosa and can promote intestinal absorption of nutrients,which may be an important mechanism for acupuncture to affect intestinal microecology.
