Study On The Association Between TGF-β2 Gene And VDR Gene Polymorphism About Resistance Exercise Effect In Ordinary People

Abstract/Summary:
Research background:The study of molecular genetic markers has been widely used in sports science,which not only improves the scientific nature of sports material selection,but also opens up a new way for the research of personalized exercise prescription.Objective:To analyze the association between VDR,TGF-β2 gene polymorphism and resistance training intervention effect in Chinese Han population,so as to screen out the molecular markers that are more sensitive to resistance training intervention,and its significance is to provide meaningful molecular genetic markers for the formulation of individualized and precise resistance exercise prescription for the public.Research Methods:191 domestic adults without exercise habit were selected as subjects,aged 18-30,without exercise contraindications,and without systematic training experience in recent three months.Squat and bench press anti-resistance intervention was conducted.Intervention scheme:10 times*5 groups*70%1RM,2 minutes interval between the groups,twice a week for 12 weeks.Before and after the intervention,the subjects’ squat and bench press 1RM,constant speed and length pedal strength,Dexa,CMJ,rectus femoris muscle thickness and pectoralis major muscle thickness were measured.At the same time,peripheral blood DNA was extracted.VDR and TGF-β2 gene polymorphisms were classified by gene chip scanning.The association between the selected loci polymorphisms and the effect of resistance training was analyzed by comparing each genotype group before and after intervention and multiple comparisons between groups.The subjects with effect quantity lower than 0.2 were treated with intervention after changing the regimen(5times*5groups*85%1RM,8 weeks),and the association between the selected sites and the effect of resistance training was verified again.Results:The polymorphism of VDR gene rs7975232 site showed no significant difference in the rate of change of each indicator between the groups.After changing the intervention program,the change rate of lower limb muscle mass in the combined AA and AC genotype group was significantly higher than that in the CC genotype group(P<0.05).In comparison between baseline and post-intervention groups,the pectoralis major muscle strength and isolength pedal muscle strength of type ID were significantly higher than those of type Ⅱ genomes(P<0.05).In comparison between groups of change rate,the squat 1RM and rectus femoris muscle thickness of type ID were significantly lower than those of type Ⅱ genomes(P<0.05).In male,the change rate of rectus femoris muscle thickness was significantly lower than that of genotype Ⅱ(P<0.05).The variation rate of squat 1RM in females with DD type was significantly higher than that with ID and Ⅱ type genomes(P<0.05).After changing the intervention program,the change rate of 1RM of genomic squat of DD type in male and female was significantly lower than that of type Ⅱ and ID combined group(P<0.05).Research Conclusions:1.The polymorphism of VDR gene rs7975232 was not associated with resistance training sensitivity of medium load(70%1RM),but was associated with resistance training sensitivity of medium and high load(85%1RM),suggesting that A allele could be used as A molecular marker to develop resistance exercise prescription sensitivity of medium and high load(85%1RM)in Chinese Han.2.Polymorphism at rs10482719 of TGF-β2 gene was associated with susceptibility to moderate load(70%1RM)resistance training,and there were differences between men and women.Prompt type Ⅱ(insert)gene and/or the I(insert)alleles can be used as the han Chinese men medium load(70%1 rm)resistance exercise prescription sensitivity molecular markers,the DD(missing)gene and/or D(missing)alleles can be used as han Chinese women in China medium load(70%1 rm)resistance molecular markers of the sensitivity of the exercise prescription.
