Abstract/Summary:
The artificial intestine digestive system is a set of instruments for simulating the intestinal digestion process in vitro by simulating the physiological conditions of various parts of the human intestine.Because of the research on human intestinal digestion,there have been outstanding problems such as sampling difficulties,high cost and ethical ethics,which hinders researchers from closely related to human health,such as food nutrition,intestinal digestion,and intestinal micro-ecology.research process.The artificial intestine digestive system has the advantages of convenient sample extraction,digestive process "visible",more economical and no ethical issues,and has broad application prospects in food nutrition and human intestinal digestion process.In this study,through the use of five continuous fermenters and a computer software-based adjustment control module,the human intestinal digestive environment was simulated to some extent,meeting the conditions for carrying out in vitro digestion experiments.The artificial intestinal digestive system was mainly studied from the following aspects::(1)Build the hardware required for the artificial intestinal digestive system.In this part,we focus on the reaction bottle of the system,the functional interface connection method is designed,and the automatic sample introduction system and the pH automatic control system are designed for the problems of large labor volume and complicated operation of the digestion experiment.The former realizes the digitization and automation of various feed liquid transfer in the experiment,the latter realizes the real-time monitoring and automatic maintenance of the pH value of the culture liquid.In the research,the innovative PID control algorithm is used in the self-designed pH value automatic regulation.In the system,the problem of hysteresis of pH measurement is solved,and the precise adjustment of pH value is realized.(2)Reliability tests were carried out on each functional module of the designed in vitro digestive system.In this part,the designed sealed reaction bottle was tested for sealing performance to meet the expected design requirements.The digital accuracy of the timing start,quantitative and fixed speed feed liquid of the automatic sample introduction system was tested.The result shows the timing success rate is 100%,the fixed speed and quantitative addition digital accuracy is above 95%,in line with the accuracy requirements of ordinary instruments;the pH automatic control system has been tested,the results show that in the test test for up to 13 hours,the pH is controlled Within the set target range;temperature control and anaerobic modules were also tested,which met the requirements of in vitro digestion experiments.(3)The human intestinal flora was cultured in this system,and a simulated digestion experiment was carried out.By using 16S rDNA identification,the time required for the intestinal flora to reach a stable phase in the system was determined.Based on the results of conventional 16S rDNA identification method,the method of using the electronic nose to detect the upper gas of the culture medium in the reaction flask can be used to determine the stability of the intestinal flora.The experimental results show that the method has high Feasible.This method is expected to solve the problem that the 16S rDNA identification is used to judge the stable period of the colony growth,and the output cycle is too long and expensive.
