Cancer,the first killer,that is seriously threatening life and health of human being.Cancer patients who died from transformation and diffusion of cancer cells is over 90%of the whole cancer group.Circulating tumor cells(CTCs)are those tumor cells moving in blood or lymph system after dropping off from tumor in situ,and infiltrating surrounding orgnization.Blood circulation is an significant approach of tumor transformation to liver,lung,brain and bone,etc.But,relative quantity of CTCs in blood is extremly small.Burying in such quantitative normal cells,CTCs in such extremly low density are difficult to be accurately captured.This thesis provides magnetic seperation targetting cancer cells in order to capturing and eliminating CTCs,expecting increasing 5-year surviving rate.The research detail is as following:(1)Calculating absorption energy between several materials and Fe3O4give the results of sodium alginate,Chitosan oligosaccharide(OCS),polyethylene glycol(PEG)have acceptable absorption energy,such as-39.23kcal/mol of sodium alginate,-43.14kcal/mol of OCS and-26.86kcal/mol of PEG,There is autotative absorption balance.Calculating their(OCS,PEG,sodium alginate)compatibility with lipid molecules resulted OCS,PEG,sodium alginate have acceptable compatibility.Calculating toxicity of these three resulted that they are such materials with excellent hydrophilicity,good solubility and low toxicity.(2)PEG,sodium alginate and OCS can pack Fe3O4by different methods,and result in different suspension effect.PEG-Fe3O4by hydrothermal method dispersing in such dispersant(PEG adding sodium oleate)that has the greatest suspension for several days with particle diameter of 80 nm.The second is OCS-Fe3O4by co-precipitation in situ with 145 nm diameter and suspension for several hours.Sodium alginate-Fe3O4has the shortest suspension time with the largest diameter.At last,PEG-Fe3O4was chosen to enter into cell experiments.(3)Red cell membrane(RBC)of 14 mg/m L was obtained through hypo-tonic destroying of membrane method combining with high speed centrifugation.Taurocholic acid sodium decorated RBC by the weight proportion of 1:4.RBC was softest in 38。C~45。C.RBC was co-decorated by targeting molecule,folic acid,and fluorescent molecule FITC.Every mg membrane can absorb 0.012 mg FITC and 0.018 folic acid.PEG-Fe3O4was loaded into red cell membrane extruded through membrane by high pressure resulting targeting fluorescent complex of PEG-Fe3O4-RBC.This thesis provided a magnetic core Fe3O4and complex particle by Fe3O4-scattering materials.Red blood cell membrane was decorated by foliac acid and fluorescein isothiocyanate isomer to a fluorescein tagetting RBC carrier.Fe3O4complex particles were packed into RBC.The products were highly dispersive magnetic particles.Experimental research showed that these particles had excellent selectictive absorption to cancer cells,sensitive magnetic response and thermal-optic effect,as well as independence from immune cells.The whole manufacture process had low cost,simple and environment-friendly progress.It is benefit to be industrialized.
