| Distant hybridization is one of the effective method to create new species and new varieties,which has been widely used in the rice breeding in China.However,there is still a lack of theoretical basis and credible experimental evidence for distant hybridization.This study analyzed whether exogenous DNA of Astragalus membaranaceu was introduced into the distant hybrid,compared the antioxidant activities and related components of red rice and its receptor rice(white),and studied molecular basis of red pericarp of red rice using distant hybrid F1 between A.embaranaceus(male)and Oryza sativa(female)and their inbred lines of red rice as materials.The results showed that one of the tested plants of F1 generation contained the SRAP marker of A.embaranaceus,which was successfully transformed into the specific SCAR marker and applied to the assisted selective breeding of the distant hybrid between A.embaranaceus and O.sativa.Red rice,containing the specific SCAR marker of A.embaranaceus,showed higher antioxidant activity and higher contents of total flavonoids and procyanidins than white rice.The Rc gene and Rd gene controlling the red pericarp in red rice both showed a transversion,causing missense mutation and reverse mutation,respectively,which may be the molecular basis red pericarp in red rice hybrid.These results proved the feasibility of distant hybridization between A.embaranaceus and O.sativa. |
